Abstract

Oksigenselainsebagaisenyawa gas terpentinguntukmelaksanakanfungsimetabolismejugadapatdigunakansebagaiterapi .Terapioksigenmerupakanterapitambahan yang dapatdimanfaatkanuntukbeberapasituasiklinisseperti hypoxemia, keracunankarbonmonoksida, trauma, dan ischemia.Administrasioksigendengankonsentrasitinggidanwaktu yang lama dapatmenyebabkan stress oksidatif. Rosella ungu (Hibiscus sabdariffaL.) diketahuimengandungantosianin yang dapatmenurunkan ROS danradikalbebasdalamtubuh.Tujuanpenelitianiniadalahuntukmengetahuipengaruhp emberianekstrak rosella ungupadamencit yang diinduksioksigenberlebihterhadapkadarmalondialdehida (MDA) dangambaranhistopatologi organ otakpadamencitputih(Musmusculus). Hewan coba yang digunakan adalah Mencit putih jantan dengan usia 8-12 minggu dengan berat 20-30 gram. Mencit dibagi menjadi 5 kelompok yaitu kelompok kontrol negatif, kelompok kontrol positif dengan paparan oksigen dengan konsentrasi FiO2 0,8 yang diberikan selama 8 jam dalam 7 hari, kelompok terapi dosis 0,378 mg/grBB0,756 mg/grBB, 1,134 mg/grBBekstrak rosella ungu. Ekstrak rosella ungu (Hibiscus sabdariffa L.) diberikansecarasondelambung 1 kali sehariselama 14 hari.Kadar MDA dilakukandenganuji TBA dandianalisisdenganone wayANOVAdandilanjutkandenganujiTukeydengan α = 1%danhistopatologiotakdenganpewarnaan HE yang dianalisisdandisajikansecaradeskriptif. Hasilpenelitianmenunjukkanbahwapemberianekstrak rosella ungudengandosis 1,134 mg/grBBmerupakandosisyang mampumenurunkankadarmalondialdehidasangatnyata(p < 0,01) denganpersentasetertinggiyaitusebesar 37,85% darikontrolpositifdandapatmemperbaikihistopatologipadasel neuron dan neuroglia ditandaidenganberkurangnyanekrosis sel.Kesimpulandaripenelitianiniyaituantosianinpadaekstrak rosella ungudapatmenurunkankadar MDA danmemperbaikigambaranhistopatologiotakmencit yang diinduksioksigenberlebih.

English Abstract

Oxygen apart as the most important gas compound to carry out metabolic functions can also be used as a therapy.Oxygen therapy is an additional therapy that can be utilized for several clinical situations such as hypoxemia, carbon monoxide poisoning, trauma, and ischemia. High-concentration oxygen administration and long periods of time can cause oxidative stress.Purple rosella (Hibiscus sabdariffa L.) is known to contain anthocyanin which can reduce ROS and free radicals in the body.The purpose of the study is knowing about the effect of purple rosella extract on mice that induced high dose oxygen into the level of MDA and the Histopathology imaging of Mice brain tissue (Musmusculus). The Tested animals must be male, 2-3 months old,with body weight 20-30 gram.Mice were divided into 5 groups: negative control group, positive control group with presentation of 0.8 FiO2 oxygen given for 8 hours in 7 days, therapy group dose 0.378 mg / grBB, 0.756 mg / grBB, 1.134 mg / grBB purple roselle extract.Purple roselle extract (Hibiscus sabdariffa L.) was given in gastric sonde once a day for 14 days.MDA levels were carried out by TBA test and analyzed by one way ANOVA and followed by Tukey test with α = 1% and the histopathology imaging analyzed with HE stain that would be observed and descripted. The results showed that the administration of purple roselle with a dose of 1.134 mg / grBB was significantly different ( p < 0,01) able to reduce malondialdehyde levels with the highest percentage of 37.85% from positive controlsand could repair neurons and neuroglia cell histopathology characterized by reduced cell necrosis.The conclusion of this study is that anthocyanin in purple roselle extract can reduce MDA levels and improve the histopathological imaging of excess oxygen-induced brain mice.
