Abstract

Glomerulonefritis merupakan penyakit ginjal karena adanya inflamasi dan proliferasi sel glomerulus, ditandai oleh peningkatan ekspresi Interleukin 1 beta (IL-1β) dan peningkatan jumlah sel radang. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak etanol Blueberry pada kasus Glomerulonefritis Akut (GNA) terhadap ekspresi Interleukin-1 beta (IL-1β) dan jumlah sel radang. 20 ekor tikus jantan strain wistar yang berumur 6-8 minggu dengan berat badan 150-250 gram dibagi dalam 5 kelompok perlakuan yang terdiri dari kontrol negatif, kontrol positif (diinduksi streptokinase sebanyak 3 kali dosis 6000 IU/ekor dengan interval 5 hari secara IV), perlakuan 1,2,3 diinduksi streptokinase sebanyak 3 kali dosis 6000 IU/ekor dengan interval 5 hari secara IV dan pemberian terapi ekstrak etanol Blueberry dengan dosis masing-masing 500 mg/kg BB, 1000 mg/kg BB, 1500 mg/kg BB selama 14 hari. Setelah 30 hari, ginjal tikus diambil untuk pemeriksaan ekspresi IL-1β menggunakan pewarnaan imunohistokimia dan jumlah sel radang menggunakan pewarnaan hematoksilin eosin, kemudian dianalisa secara statistic menggunakan One Way ANOVA dengan taraf kepercayaan sebesar 95% atau α = 0,05. Hasil penelitian menunjukkan pemberian ekstrak etanol Blueberry dapat menurunkan ekspresi IL-1β dan jumlah sel radang pada organ ginjal dengan dosis terbaik yaitu 1500 mg/kg BB. Kesimpulan dari penelitian ini adalah ekstrak etanol Blueberry memiliki efek terapi terhadap glomerulonefritis akut pada tikus putih yang diinduksi streptokinase berdasarkan ekspresi IL-1β dan jumlah sel radang.

English Abstract

Glomerulonephritis is a kidney disease which is indicated by inflammation and proliferation glomerular cell characterized by an increase in the expression of Interleukin 1 beta (IL-1β) and number of inflammatory cells. The aim of this study is to ascertain the effect of Blueberry ethanol extract on cases of Acute Glomerulonephritis (GNA) on Interleukin-1 beta (IL-1β) expression and increasing of total inflammatory cells. 20 wistar male rats aged 6-8 weeks with weight of 150-250 grams were divided into 5 treatment groups consisting of negative control, positive control (induced by streptokinase 3 times with dose 6000 IU/each with interval 5 days, IV), three groups of treatments induced by streptokinase 3 times with dose 6000 IU/each with interval 5 days and treatments of Blueberry ethanol extract with dose 500 mg/kg BW, 1000 mg/kg BW, 1500 mg/kg BW for 14 days, respectively. After 30 days, rat kidneys were taken for examination of IL-1β expression was using immunohistochemistry and total of inflammatory cells with Hematoxilin-Eosin staining, then analyzed statistically using One Way ANOVA with a confidence level of 95% or α = 0.05. The results showed that the administration of Blueberry ethanol extract as therapy could reduce IL-1β expression and number of inflammatory cells of kidney organ with the best dose of 1500 mg/kg BW. The conclusion of this research is that Blueberry ethanol extract has a therapeutic effect on acute glomerulonephritis that cause by streptokinase based on levels of IL-1β expression and increasing of total inflammatory cells.
