Abstract

Hiperkolesterolemia adalah peningkatan kadar kolesterol dalam darah. Pencegahan peningkatan kadar trigliserida dan kerusakan jaringan lambung akibat hiperkolesterolemia dapat dengan memberikan arang aktif. Interaksi antara kolesterol dengan karbon aktif terjadi adsorpsi secara fisika. Penelitian ini bertujuan untuk mengetahui pengaruh terapi preventif arang aktif terhadap kadar trigliserida dalam tubuh dan histopatologi lambung tikus (Rattus norvegicus) yang diinduksi hiperkolesterol. Penelitian ini menggunakan tikus putih (Rattus norvegicus) strain Wistar dibagi dalam 6 kelompok perlakuan yaitu: Kelompok negatif, kelompok positif diberi diet hiperkolesterolemia sebanyak 3,02 g/ekor, kelompok P4 diberikan arang aktif sebanyak 680 mg/ekor, kelompok perlakuan diberi diet hiperkolesterolemia sebanyak 3,02 g/ekor dan terapi arang aktif 340 mg/ekor, 680 mg/ekor, dan 1020 mg/ekor diberikan peroral mulai hari ke 1-14 hari. Kadar trigliserida diamati dengan metode spektrofotometri, dianalisa dengan uji ANOVA dilanjutkan uji Tukey (p<0,01). Gambaran histopatologi lambung diamati secara kualitatif dengan pewarnaan HE (hematoxylin and eosin).Hasil penelitian menunjukkan bahwa pemberian preventif arang aktifmampu mencegah peningkatan kadar trigliserida dan dapat mencegah kerusakan gambaran histopatologi lambung. Dosis680 mg/150 gBBmerupakan dosis terbaik yang mampu menunjukkan hasil mendekati kelompok Kontrol Negatif.Kesimpulan dari penelitian ini adalah arang aktif dapat digunakan sebagai preventif pada tikus putih (Rattus norvegicus) model hiperkolesterolemia.

English Abstract

Hypercholesterolemia is an increase of cholesterol levels in the blood. Prevention of increased triglyceride levels and damage to gastric tissue due to hypercholesterolemia can be conducted by providing activated charcoal. The interaction between cholesterol and activated carbon is physically adsorbed. This study aimed to determine the preventive effect of activated charcoal of triglyceride levels in the body and hypercholesterol-induced gastric histopathology of rats (Rattus norvegicus) induced by hypercholesterol. This study used Wistar strain of Rattus norvegicus that divided into 6 treatment groups, such as: Negative group, positive group given hypercholesterolemic diet as much as 3.02 g/rat, P4group with activated charcoal 680 mg/rat, treatment group given a hypercholesterolemic diet as much as 3.02 g/rat and activated charcoal therapy 340 mg/rat, 680 mg/rat, and 1020 mg/rat were given orally starting from 1-14 days. Triglyceride levels were observed by spectrophotometric method, analyzed by ANOVA test and follow by Tukey test (p <0.01). Gastric histopathology was observed qualitatively with HE staining (hematoxylin and eosin). The results showed that preventive activated charcoal was able to prevent increase of triglyceride level and prevent damage to gastric histopathology. The dose of 680 mg / 150 g BB was the best dose that is able to show the results of approaching the negative control group. The conclusion of this study was that activated charcoal can be used to prevent hypercholesterolemic in rat (Rattus norvegicus).
