Abstract
Fibrosis hati adalah perubahan komposisi dan peningkatan ECM yang terjadi akibat penyembuhan cedera secara kronik. Salah satu regulasi remodeling ECM diperankan oleh TIMP-1, sebagai penghambat degrasi ECM, yang merupakan peran utama TIMP-1 dalam fibrogenesis. Penelitian ini bertujuan untuk menguji pengaruh ekstrak etanol daun Moringa oleifera (MO) dalam menurunkan ekspresi TIMP-1 jaringan hati pada tikus model fibrosis hati. Metode yang digunakan adalah Randomized posttest only controlled group design. Penelitian ini menggunakan 30 sampel hewan uji yaitu tikus putih (Rattus novergicus strain wistar) jantan yang dibagi menjadi 5 kelompok. Kelompok tanpa perlakuan yang diberikan NaCl, kelompok 2 diinjeksi CCl4 (1 mL/KgBB dengan perbandingan CCl4 dan minyak jagung 1:9, i.p. 2x seminggu selama 12 minggu dilanjutkan dengan 2 mL/KgBB selama 13 hari). Ke-3 kelompok lainnya, tikus diinjeksi dengan dosis yang sama untuk CCl4 dan diberikan dosis daun Moringa oleifera yang berbeda (150 mg/KgBB 300 mg/KgBB, 600 mg/KgBB) setiap hari selama 97 hari. Pengukuran ekspresi TIMP-1 dengan menghitung jumlah sel yang mengekspresian TIMP-1 jaringan hati pada hasil pemfotoan immunofluorescence. Kelompok kontrol positif yang hanya diberikan CCl4 memiliki ekspresi TIMP-1 yang tertinggi, kelompok tanpa perlakuan memiliki ekspresi TIMP-1 yang terendah. Uji ANOVA menunjukkan adanya perbedaan ekspresi TIMP-1 yang signifikan antara ke-5 kelompok tikus. Hasil dari perbandingan antara kelompok yang diinduksi CCl4 (K-Pos) dengan masing-masing ke-3 kelompok perlakuan adalah tidak signifikan. Dengan demikian, dapat disimpulkan bahwa pemberian ekstrak etanol daun Moringa oleifera tidak dapat menurunkan ekspresi TIMP-1 jaringan hati pada tikus model fibrosis hati. Grafik korelasi yang menunjukkan nilai signifikansi 0,000 dengan koefisien -0.534 yang masuk dalam rentang cukup kuat. Hal tersebut menyimpulkan bahwa Ekspresi TIMP-1 semakin menurun seiring dengan penambahan dosis ekstrak etanol daun Moringa oleifera yang diberikan.
English Abstract
-Liver fibrosis is a change in composition and an increase in ECM that occurs as a result of healing chronic injuries. One of the ECM remodeling regulations is played by TIMP-1, as an inhibitor of ECM degradation, which is the main role of TIMP-1 in fibrogenesis. This study aims to examine the effect of ethanol extract of Moringa oleifera (MO) leaves in reducing the expression of TIMP-1 liver tissue in mice with liver fibrosis. The method used is randomized posttest only controlled group design. This study used 30 samples of test animals namely male rats (Rattus novergicus strain wistar) divided into 5 groups. The untreated group was given NaCl, group 2 was injected with CCl4 (1 mL / KgBB with a ratio of CCl4 and corn oil 1: 9, i.p. 2x a week for 12 weeks followed by 2 mL / KgBB for 13 days). The other 3 groups, mice were injected with the same dose for CCl4 and given different doses of Moringa oleifera leaves (150 mg / KgBB 300 mg / KgBB, 600 mg / KgBB) every day for 97 days. TIMP-1 expression measurement by calculating the number of cells expressing TIMP-1 liver tissue in immunofluoerescence photo results. The positive control group which was only given CCl4 had the highest TIMP-1 expression, the treatment group had the lowest TIMP-1 expression. ANOVA test showed a significant difference in TIMP-1 expression between the 5 groups of mice. The comparison between groups induced by CCl4 (K-Pos) with each of the 3 treatment groups are not significant. Thus, it can be concluded that the administration of ethanol extract of Moringa oleifera leaves cannot reduce the expression of TIMP-1 liver tissue in mice with liver fibrosis. The correlation chart shows a significance value (0,000) with a coefficient -0,534 which means strong enough. It was concluded that TIMP-1 expression decreased with increasing doses of ethanol extract from Moringa oleifera leaves.
