Abstract
Toxoplasma gondii merupakan parasit patogen intraseluler yang dapat memiliki kemampuan untuk menginfeksi semua sel berinti dan diduga menyebabkan obesitas. Profilin merupakan komponen alat gerak Toxoplasma gondii yang dapat meningkatkan interleukin-12 (IL- 12) sehingga menyebabkan inflamasi adiposa dan menyebabkan obesitas. Tujuan penelitian ini adalah untuk mengetahui efek paparan profilin Toxoplasma gondii terhadap hipertrofi dan hiperplasia adiposit pada tikus yang diberi diet hiperkalori. Studi bersifat eksperimental menggunakan post-test only terhadap tikus Wistar jantan selama 16 minggu. 65 ekor tikus terbagi 13 kelompok perlakuan. Variabel yang diukur adalah ukuran dan jumlah adiposit menggunakan metode HE. Pengujian menggunakan uji ANOVA, korelasi Pearson, dan uji regresi linear. Didapatkan hasil penelitian pada uji ANOVA 0,111 dan 0,781 yang artinya tidak ada perbedaan pengaruh dosis profilin terhadap hipertrofi dan hiperplasia. Hasil regresi linear untuk injeksi satu kali memiliki rata-rata nilai p > 0,05 yang diberi diet normal maupun diet hiperkalori yang tidak signifikan, berkorelasi sangat lemah dan berlawan. Pada hasil diinjeksi dua kali memiliki nilai ratarata p > 0,05 yang tidak signifikan. Pada delta berat badan nilai ANOVA 0,004 artinya ada perbedaan pengaruh dosis profilin Toxoplasma gondii. Dapat disimpulkan bahwa profilin Toxoplasma gondii berpengaruh terhadap berat badan, tetapi tidak mempengaruhi ukuran dan jumlah adiposit.
English Abstract
Toxoplasma gondii is an intracellular pathogenic parasite that can infect virtually any nucleated cells and is thought to cause obesity. Profilin is a component of Toxoplasma gondii which can increase interleukin-12 (IL-12), which may lead to adipose inflammation and causes obesity. This study was aimed at determining the effect of Toxoplasma gondii profilin exposure on hypertrophy and hyperplasia in rats given a hypercaloric diet. This experimental study used posttest only design applied to male Wistar rats for 16 weeks. 65 rats were divided into 13 treatment groups. The variables measured were the size and number of adipocytes using the HE method. The data were analyzed using ANOVA, Pearson correlation, and linear regression tests. The results obtained from the ANOVA test were 0.111 and 0.781, which means there was no difference in effect of dose of profilin on hypertrophy and hyperplasia. The linear regression results for one-time injection showed a mean value of p> 0.05, which means there was no significance difference between the group given a normal diet and the group with hypercaloric diet, and the correlation was very weak and negative. The result of two injections had an average value of p> 0.05, which was not significant. The delta weight ANOVA test resulted 0.004, which means that there were differences in the effects of the dose of profilin Toxoplasma gondii. It can be concluded that profilin Toxoplasma gondii affects body weight, but has no effect on the size and number of adipocytes.
