Abstract

Antibiotik merupakan senyawa alami atau senyawa sintetik yang dapat menghambat serta membunuh aktivitas bakteri.Kasus resitensi antibiotik merupakan masalah global yang banyak terjadi baik pada negara maju dan berkembang seperti Indonesia. Seiring meningkatnya resistensi antibiotik, banyak dilaukan penelitian untuk mendapatkan senyawa baru yang berasal dari bahan alam, salah satunya buah Prunus persica Zieb & Zucc yang banyak digunakan masyarakat Bromo Tengger Semeru dalam mengobat berbagai penyakit. Penelitian ini bertujuan untuk mengetahui profil metabolit sekunder dan aktivitas antibakteri ekstrak etanol 80% , fraksi kloroform, fraksi etil asetat, fraksi n-butanol dan fraksi air terhadap bakteri Streptococcus pneumoniae. Pengujian sampel terdiri dari beberapa tahapan yaitu ekstraksi dilakukan dengan cara maserasi menggunakan pelarut etanol 80%, identifikasi fitokimia menggunakan metode Kromatografi Lapis Tipis (KLT) dan diteruskan dengan pengujian aktivitas antibakteri menggunakan metode mikrodilusi untuk menentukan nilai Kadar Hambat Minimal (KHM) dan nilai Kadar Bunuh Minimal (KBM). Hasil penelitian menunjukkan dalam fraksi-fraksi Buah (Prunus persica Zieb &Zucc) mengandung flavonoid,terpenoid, antrakuinon dan polifenol. Hasil yang diperoleh dari KHM dari setiap sampel yaitu fraksi kloroform sebesar 97.66 ppm, fraksi etil asetat sebesar 390.63 ppm, fraksi n-butanol sebesar 781.25 ppm, fraksi air sebesar 50000 ppm dan ekstrak etanol tidak teramati pada konsentrasi 50000 ppm. Nilai KBM fraksi kloroform sebesar 1562.5 ppm, fraksi etil asetat sebesar 3125 ppm, fraksi nbutanol sebesar 37500 ppm, fraksi air sebesar 50000 ppm.

English Abstract

Antibiotics are natural or synthetic compounds that can inhibit and kill activities of bacterial. Antibiotic resistance is global major problem in many developed and developing countries such as Indonesia. As antibiotic resistance increases, a lot of researches has been done to get new antibiotic compounds from natural ingredients. Prunus persica Zieb & Zucc fruit is one of natural ingredients which is widely used by the Bromo Tengger Semeru people in treating various diseases. This study aims to determine the secondary metabolites profile and antibacterial activity of 80% ethanol extract, chloroform fraction, ethyl acetate fraction, n-butanol fraction and water fraction of Prunus persica Zieb & Zucc against Streptococcus pneumoniae. The sample was tested by several stages. The first stage was extraction by maceration using 80% ethanol solvent. Phytochemicals identification was done tested by Thin Layer Chromatography (TLC) method and antibacterial activity was tested by the microdilution method to determine the value of Minimum Inhibitory Concentration (MIC) and Minimal Killing Concentraon (MKC). The results showed that Fractions in Fruits (Prunus persica Zieb & Zucc) contained flavonoids, terpenoids, anthraquinones and polyphenols. MIC value of chloroform fraction was 97.66 ppm, ethyl acetate fraction was 390.63 ppm, n-butanol fraction was 781.25 ppm, water fraction was 50000 ppm and ethanol extract was not observed at concentration of 50000 ppm. The MKC value of chloroform fraction was 1562.5 ppm, the ethyl acetate fraction was 3125 ppm, the n-butanol fraction was 37500 ppm, the water fraction was 50000 ppm
