Abstract
Konsumsi MSG melebihi batas aman dapat menyebabkan terbentuknya radikal bebas akibat over aktifasi reseptor glutamat. Jika jumlah radikal bebas dan antioksidan endogen tidak seimbang akan menyebabkan stres oksidatif yang mengakibatkan kematian sel-sel tubuh termasuk nucleus arkuata di Hipotalamus. Kerusakan pada nukeleus arkuata menyebabkan gangguan produksi hormon reproduksi termasuk estrogen. Hormon estrogen berperan dalam pembentukan pembuluh darah arteriol di endometrium. Jika sekresi estrogen menurun maka pembentukan pembuluh darah arteriol tidak optimal sehingga menyebabkan gangguan saat menstruasi yaitu menorrhagia. Efek radikal bebas dalam tubuh dapat dicegah dengan pemberian antioksidan eksogen yang diperoleh dari brokoli yang telah diteliti memiliki kandungan antioksidan flavonoid tertinggi dari sepuluh sayuran yag diteliti. Tujuan penelitian ini adalah untuk mengetahui pengaruh pemberian ekstrak etanol brokoli terhadap jumlah arteriol endometrium tikus putih wistar yang dipapar MSG. Metode penelitian ini adalah true experimental laboratorium dengan menggunakan randomized post test only control group design yang dilakukan terhadap 30 tikus putih wistar. Sampel dipilih berdasarkan kriteria yang ditetapkan dan dikelompokan menjadi Kelompok kontrol negatif (tanpa paparan ekstrak brokoli dan MSG), kontrol negatif (dipapar MSG 0,7 mg/gBB), Perlakuan [P I (MSG 0,7 mg/gBB + ekstrak brokoli 500 mg/KgBB), P II (MSG 0,7 mg/gBB + ekstrak brokoli 1000 mg/KgBB), P III (MSG 0,7 mg/gBB + ekstrak brokoli 2000 mg/KgBB)] yang diberikan per oral selama 28 hari. Jumlah arteriol endometrium dilihat menggunakan mikroskop olympus BX51 dan software OlyVIA. Analisa hasil penelitian dengan menggunakan uji Anova one way dan post hoc test menunjukan bahwa terjadi penurunan jumlah arteriol endometrium pada kelompok kontrol positif serta terdapat peningkatan jumlah arteriol pada P I, P II, P III secara bermakna. Kesimpulan dari penelitian ini adalah pemberian MSG dapat menurunkan jumlah arteriol endometrium tikus putih wistar dan pemberian ekstrak etanol brokoli dapat meningkatkan jumlah arteriol endometrium tikus putih wistar yang dipapar MSG.
English Abstract
Consumption of MSG exceeds the safe limit can cause the formation of free radicals due to over activation of glutamate receptors. If the free radicals and endogenous antioxidants is not balanced will cause oxidative stress which results in the death of body cells including the arcuate nucleus in the hypothalamus. Damage to the arcuate nucleus causes a disruption in the production of reproductive hormones including estrogen. Estrogen hormones play a role in the formation of arterioles in the endometrium. If estrogen secretion decreases, arteriole blood vessel formation is not optimal, causing menstrual disorders, namely menorrhagia. The effects of free radicals in the body can be prevented by giving exogenous antioxidants obtained from broccoli that have been studied to have the highest content of flavonoid antioxidants from ten vegetables studied. The purpose of this study was to determine the effect of broccoli ethanol extract on the number of wistar white rat endometrial arterioles exposed by MSG. This research method is true experimental laboratory using randomized post-test only control group design carried out on 30 wistar white rats. Samples were selected based on established criteria and grouped into negative control groups (without exposure to broccoli extract and MSG), negative controls (exposed to MSG 0.7 mg/gBB), Treatment [P I (MSG 0.7 mg/gBB + broccoli extract 500 mg/KgBB), P II (MSG 0.7 mg/ gBB + broccoli extract 1000 mg / KgBB), P III (MSG 0.7 mg / gBB + broccoli extract 2000 mg / KgBB)] which was given orally for 28 days. The number of endometrial arterioles was seen using the Olympus BX51 microscope and OlyVIA software. Data were analyzed using one way ANOVA and Post Hoc Tukey. Analysis of the results using the one way Anova test and post hoc test showed that there was a decrease in the number of endometrial arterioles in the positive control group and that there was a significant increase in the number of arterioles in P I, P II, P III. This study concluded that MSG can decrease the number of wistar white rats endometrial arterioles, while giving broccoli ethanol extract can increase the number of wistar white rat endometrial arterioles exposed by MSG.
