Abstract

Kedelai merupakan komoditas pangan penting di Indonesia. Berbagai persoalan banyak ditemukan diantaranya persoalan kesehatan lahan akibat tercemar herbisida oksifluorfen dan kesehatan tanaman akibat terserang penyakit rebah semai. Langkah alternatif yang dapat dilakukan yakni melakukan bioremediasi lahan dengan memanfaatkan rhizobakteri. Rhizobakteri dianggap mampu mendegradasi senyawa residu herbisida dan menghambat pertumbuhan patogen S. rolfsii. Penelitian bertujuan untuk mengkaji potensi bakteri rhizosfer dalam mendegradasi residu herbisida oksifluorfen dan menghambat pertumbuhan jamur patogen Sclerotium rolfsii pada tanaman kedelai. Penelitian dilaksanakan di Laboratorium Penyakit Tumbuhan, Rumah Kawat Fakultas Pertanian Universitas Brawijaya, dan Laboratorium Kimia Fakultas Matematika dan Ilmu Pengetahuan Alam Universitas Muhammadiyah Malang pada bulan Januari hingga Mei 2019. Penelitian menggunakan Rancangan Acak Kelompok (RAK) dengan 8 perlakuan dan 4 ulangan. Perlakuan diantaranya menggunakan satu konsentrasi herbisida oksifluorfen 2 ml/l, 6 isolat rhizobakteri terseleksi, dan jamur patogen Sclerotium rolfsii. Data kemudian dianalisis menggunakan ANOVA dengan analisis lanjutan uji DMRT dengan taraf 5%. Hasil penelitian menunjukkan bahwa uji kemampuan hidup bakteri pada media NA mengandung herbisida oksifluorfen dengan konsentrasi 2 ml/l terdapat 10 isolat rhizobakteri yang mampu tumbuh antara lain isolat Bacillus sp. (N3), Clostridium sp. (N10), Xanthomonas sp. (N23), Pseudomonas sp. (N24), Pantoea sp. (N26), Erwinia sp. (N27), Pseudomonas sp. (H13), Clostridium sp. (TH16), Erwinia sp. (TH26), Pantoea sp. (TH37). Pada uji penghambatan pertumbuhan jamur patogen Sclerotium rolfsii terdapat 6 isolat rhizobakteri yang mampu menghambat antara lain isolat Bacillus sp. (N3), Clostridium sp. (N10), Xanthomonas sp. (N23), Pseudomonas sp. (N24), Pantoea sp. (N26), Erwinia sp. (N27). Enam (6) isolat rhizobakteri tersebut juga mampu menekan penyakit rebah semai tanaman kedelai dalam skala rumah kawat dibandingkan kontrol pada 6, 9, dan 12 hari setelah inokulasi. Pada uji potensi bakteri dalam mendegradasi herbisida oksifluorfen terhadap pertumbuhan tanaman kedelai dalam skala rumah kawat menunjukkan bahwa 6 isolat rhizobakteri memiliki kemampuan memacu pertumbuhan tanaman kedelai pada variabel tinggi tanaman, jumlah daun, dan panjang akar pada 24 hari setelah tanam. Pada uji residu oksifluorfen setelah aplikasi isolat rhizobakteri menunjukkan bahwa isolat Pseudomonas sp. (N24) mampu mengurangi residu oksifluorfen hingga 84.53%, sedangkan isolat Erwinia sp. (N27) mampu mengurangi residu oksifluorfen hingga 46.10% pada 70 hari setelah inokulasi dengan menggunakan metode HPLC (High Pressure Liquid Chromatography).

English Abstract

Soybean is an important food commodity in Indonesia. Many problems have been found including the health problems of the land due to contamination of oxyfluorphene herbicide and plant health due to seedlings. An alternative step that can be done is to do land bioremediation using rhizobacteria. Rhizobacteria are thought to be able to degrade herbicide residual compounds and inhibit the growth of Sclerotium rolfsii pathogens. The study aimed to examine the potential of rhizosphere bacteria in degrading residual oxyfluorphene herbicide and inhibiting the growth of Sclerotium rolfsii pathogenic fungi on soybean plants. The research was conducted at the Laboratory of Plant Diseases, Glass Houses of the Faculty of Agriculture, University of Brawijaya, and Chemistry Laboratory of the Faculty of Mathematics and Natural Sciences University of Muhammadiyah Malang in January to May 2019. The study used a Randomized Block Design (RBD) with 8 treatments and 4 replications. The treatments included using a concentration of 2 ml / l of oxyfluorphene herbicides, 6 selected rhizobacterial isolates, and Sclerotium rolfsii pathogenic fungi. Data were then analyzed using ANOVA with further analysis of the DMRT test with a level of 5%. The results showed that the test of life ability on NA media contained oxyfluorphene herbicide with 2 ml/l of concentration there were 10 rhizobacterial isolates that were able to grow, including Bacillus sp. Isolates. (N3), Clostridium sp. (N10), Xanthomonas sp. (N23), Pseudomonas sp. (N24), Pantoea sp. (N26), Erwinia sp. (N27), Pseudomonas sp. (H13), Clostridium sp. (TH16), Erwinia sp. (TH26), Pantoea sp. (TH37). In the test of the inhibition of the growth of Sclerotium rolfsii pathogenic fungi, 6 isolates of rhizobacteria were able to inhibit, among others, Bacillus sp. Isolates. (N3), Clostridium sp. (N10), Xanthomonas sp. (N23), Pseudomonas sp. (N24), Pantoea sp. (N26), Erwinia sp. (N27). The six (6) isolates of rhizobacteria had the ability to suppress soybean seedling disease in the glass house scale compared to control at 6, 9, and 12 days after inoculation. In the test of the potential of bacteria to degrade oxyfluorphene herbicide on soybean plant growth in the glass house scale showed that 6 rhizobacterial isolates had the ability to spur soybean growth on variable plant height, leaf number, and root length 24 days after planting. In the oxyfluorphene herbicide residue test after the application of rhizobacterial isolates showed that Pseudomonas sp. (N24) can reduce oxyfluorphene herbicide residues by up to 84.53%, while Erwinia sp. Isolates. (N27) can reduce oxyfluorphene herbicide residues up to 46.10% at 70 days after inoculation using the HPLC (High Pressure Liquid Chromatography) method.
