Abstract

Tungau Acarus siro L. (Acari: Acaridae) merupakan hama gudang yang sering disebut sebagai tungau tepung. A. siro tersebar luas di dunia dan menyerang produk-produk penyimpanan. Usaha pencegahan serangan A. siro diperlukan untuk mempertahankan kualitas dari tepung sehingga kebutuhan konsumen tercukupi. Pengembangan pengendalian dengan memanfaatkan kombinasi entomoacaripatogen Beauveria bassiana Bals. (Hypocreales: Cordycipitaceae) dengan berbagai kerapatan dan pestisida nabati ekstrak daun paitan (EDP) Tithonia diversifolia Hemsl. (Asterales: Asteraceae) dengan berbagai konsentrasi masih terbatas. Sinergisme antara cendawan entomo-akaripatogen B. bassiana dan EDP diharapkan mampu terjadi sehingga dapat meningkatkan mortalitas imago, mengurangi produksi telur dan produksi larva A. siro. Penelitian dilaksanakan pada bulan Februari hingga bulan Agustus 2018 di Laboratorium Hama Tumbuhan (HT) dan Laboratorium Pengendalian Hayati (PH) Jurusan Hama dan Penyakit Tumbuhan (HPT) Fakultas Pertanian (FP) Universitas Brawijaya (UB). Pengamatan uji kompatibilitas B. bassiana dan EDP meliputi diameter koloni, jumlah konidia dan perkecambahan konidia B. bassiana pada 6 hari setelah aplikasi (HSA) yang masing-masing dilakukan dengan 12 kombinasi perlakuan yang diulang sebanyak 4 kali sehingga didapatkan 48 satuan percobaan. Apabila hasil analisis diketahui adanya perbedaan yang nyata respon dari kombinasi jamur B. bassiana dengan EDP terhadap diameter koloni, jumlah konidia, dan perkecambahan konidia B. bassiana, maka dilanjutkan dengan uji Duncan pada taraf kesalahan 5%. Uji regresi sederhana juga dilakukan untuk mengetahui hubungan antara aplikasi EDP terhadap pertumbuhan diameter koloni, jumlah konidia dan perkecambahan konidia B. bassiana. Pengamatan uji toksisitas terdiri dari mortalitas imago A. siro, jumlah telur yang dihasilkan, jumlah larva yang dihasilkan, dan gejala infeksi yang ditimbulkan oleh aplikasi B. bassiana dan EDP terhadap morfologi imago A. siro. Hasil penelitian menunjukkan bahwa jamur entomo-akaripatogen B. bassiana kerapatan 1x104, 1x106 dan 1x108 konidia/ml aquades yang dikombinasikan dengan EDP konsentrasi 0,5; 1; dan 2% menghambat diameter koloni, sporulasi konidia, dan viabilitas konidia B. bassiana serta menunjukkan hasil yang tidak kompatibel. Pada uji regresi sederhana menunjukkan bahwa pencampuran B. bassiana dan EDP menghasilkan hubungan linear terhadap diameter koloni dan viabilitas B. bassiana serta tidak linear terhadap sporulasi konidia B. bassiana. Hasil uji toksisitas menunjukkan bahwa jumlah telur dan larva A. siro dapat ditekan menggunakan jamur B. bassiana kerapatan 1x104, 1x106 dan 1x108 konidia/ml akuades serta EDP konsentrasi 0; 0,5; 1 dan 2%. Lalu mortalitas tertinggi imago A. siro terdapat pada perlakuan B. bassiana kerapatan 1x108 konidia/ml akuades.

English Abstract

Acarus siro L. mites (Acari: Acaridae) is a storage pest which often called flour mites. A. siro is widespread around the world and attacks storage products. Prevention of A. siro is essential to ful fill the customer needs. Control development by utilizing the entomo-acaripatogen combination of Beauveria bassiana Bals. (Hypocreales: Cordycipitaceae) with various densities and paitan leaf extract (PLE) Tithonia diversifolia Hemsl. (Asterales: Asteraceae) with various concentrations are still limited. Synergism between entomo-acaripatogen B. bassiana fungi and PLE are expected to increase mortality of adult mite as well as to reduce egg production and A. siro larvae. The experiment was conducted from February to August 2018 in Laboratory of Plant Pest and Laboratory of Biological Control Department of Plant Pests and Diseases, Faculty of Agriculture, University of Brawijaya. Observation of B. bassiana and PLE compatibility tests included colony diameter, conidia number and germination of conidia B. bassiana at 6 days after application (DAA). Each of which was carried out with 12 treatment combinations which were repeated 4 times to obtain 48 experimental units. The analysis showed that the response of the combination of B. bassiana fungi and PLE were significantly different from the colony diameter, the number of conidia, and the germination of conidia B. bassiana, they will be followed by Duncan's test at the 5% error level. A simple regression test was also conducted to determine the relationship between PLE application on growth in colony diameter and number of conidia and germination of conidia B. bassiana. Observation of toxicity tests consisted of mortality of A. siro, number of eggs produced, number of larvae produced, and symptoms of infection is caused by the application of B. bassiana and PLE to the morphology of A. siro. It was found that entomo-acaripatogen B. bassiana fungi density was 1x104, 1x106 and 1x108 conidia/ml of distilled water combined with PLE concentration of 0.5; 1; and 2% inhibited colony diameter, conidial sporulation, and conidia B. bassiana viability and showed incompatible results. In a simple regression test showed that mixing B. bassiana and PLE resulted in a linear relationship to colony diameter and B. bassiana viability and was not linear to conidia B. bassiana sporulation. The experiment of the toxicity test showed that the number of eggs and A. siro larvae could be reduced by using the B. bassiana fungi density of 1x104, 1x106 and 1x108 conidia/ml of distilled water and PLE concentration of 0; 0.5; 1 and 2%. Then the highest mortality of A. siro imago was found in the treatment of B. bassiana density of 1x108 conidia/ml of distilled water.
