Abstract

Penelitian ini bertujuan untuk menganalisis pengaruh pemberian protein CSN1S2 dari susu kambing peranakan etawah terhadap sekuen gen Kir6.2 pada tikus model DMT2. Hewan coba yang digunakan adalah tikus jantan (Rattus norvegicus strain Wistar) yang dibagi menjadi dua kelompok besar, yaitu kelompok kontrol dan kelompok DMT2. Setiap kelompok diberi perlakuan berupa pemberian protein CSN1S2 375 mg/kg BB, 750 mg/kg BB, 1500 mg/kg BB, dan tanpa pemberian protein CSN1S2. Masing-masing kelompok terdiri dari tiga ulangan. DNA diisolasi dari organ otak. Gen Kir6.2 diamplifikasi menggunakan primer spesifik. Produk PCR dipurifikasi dan dilakukan sekuensing menggunakan ABI 3730xl DNA Sequencer. Sekuen DNA dianalisis menggunakan software MEGA7. DNA whole genome yang diperoleh memiliki nilai kemurnian 1,87-2,12. Konsentrasi DNA dari masing-masing sampel berkisar antara 134,83-2963,71 ng/μl. Amplifikasi gen Kir6.2 menghasilkan DNA dengan panjang 1173 bp. Tidak terdapat perubahan sekuen Kir6.2 pada seluruh perlakuan. Pembuatan tikus model DMT2 dengan terlebih dahulu mengondisikan agar terjadi hiperkolesterolemia dan memberikan 25 kg/BB streptozotocin tidak memberikan pengaruh pada gen yang berkaitan dengan homeostasis glukosa yang terdapat di otak. Penelitian ini juga memberikan hasil bahwa pemberian protein CSN1S2 dengan dosis 375 mg/kg BB, 750 mg/kg BB, dan 1500 mg/kg BB tidak menyebabkan mutasi pada gen Kir6.2 di otak tikus model DMT2.

English Abstract

The aim of this study was to analyze the effect of CSN1S2 protein of etawah crossbreed goat milk on Kir6.2 gene sequences in the rat model of T2DM. The experimental animals used were male rats (Rattus norvegicus strain Wistar) which were divided into two major groups, namely control group and T2DM group. Each group was treated by giving CSN1S2 protein 375 mg/kg BW, 750 mg/kg BW, 1500 mg/kg BW, and without giving CSN1S2 protein. Each group was replicated three times. DNA was isolated from the rat brain. Kir6.2 gene was amplified by using specific primers. PCR products were purified and sequenced by using ABI 3730xl DNA Sequencer. DNA sequences were analyzed by using MEGA7 software. The whole genome DNA had purity value of 1.86-2.12. The DNA concentration of each sample ranged from 134.83 to 2963.71 ng/μl. Amplification of the Kir6.2 gene produced 1173 bp DNA. There was no change in the Kir6.2 sequence in all treatments. The rats model of TD2M which were made to become hypercholesterolemia and given 25 kg/BB streptozotocin turned out to have no effect on genes associated with glucose homeostasis in the brain. This study also showed that administration of CSN1S2 protein at the dose of 375 mg/kg BB, 750 mg/kg BW, and 1500 mg/kg BW did not cause mutations in the Kir6.2 gene in the brain of rat model of T2DM.
