Abstract
Eugenol atau 4-allyl-2-methoxyphenol merupakan komponen utama dalam minyak cengkeh yang dapat dijadikan sebagai starting material dalam sintesis, salah satunya adalah sintesis melalui reaksi Ritter. Eugenol sebagai prekusor karbokation direaksikan dengan asetonitril, dan penambahan katalis asam kuat yaitu H2SO4 untuk membentuk senyawa organonitrogen. Jumlah mol dalam reaksi eugenol: asetonitril: H2SO4 berturut-turut sebesar 0,1: 0,5: 0,1 mol. Reaksi dilakukan pada suhu es (0oC) dengan waktu reaksi 4 jam, suhu ruang (27oC) dengan waktu reaksi 4 jam, dan suhu refluks (90oC) dengan waktu reaksi 4, 8, dan 16 jam. Hasil reaksi dikarakterisasi menggunakan spektrofotometer UV-Vis, IR, dan GC-MS. Reaksi pada suhu es (0oC) dan suhu ruang (27oC) dengan waktu reaksi 4 jam tidak menghasilkan senyawa organonitrogen. Sedangkan reaksi pada suhu refluks (90oC) dengan waktu reaksi 4 jam menghasilkan senyawa organonitrogen yaitu eugenil asetamida. Hasil reaksi pada suhu refluks (90oC) dengan waktu reaksi 8, dan 16 jam menunjukkan terbentuknya senyawa organonitrogen yaitu senyawa eugenil asetamida dan senyawa isoquinolina yang kuantitasnya semakin meningkat dengan semakin lamanya waktu reaksi. Hasil reaksi pada suhu refluks (90oC) dengan waktu reaksi 16 jam diuji sifat toksisitasnya menggunakan metode Brain Shrimp Lethality Test terhadap Artemia salina menunjukkan nilai LC50 sebesar 16 ppm dan digolongkan mempunyai aktivitas toksisitas kuat.
English Abstract
Eugenol or 4-allyl-2-methoxyphenol is the main component in clove oil which can be used as a starting material in synthesis, one of the example is synthesis through the Ritter reaction. Eugenol as a carbocation precursor is reacted with acetonitrile, and the addition of a strong acid catalyst is H2SO4 to form organonitrogen compounds. The number of moles in the reaction of eugenol: acetonitrile: H2SO4 is 0,1: 0,5: 0,1 mol. The reaction was carried out at ice temperature (0oC) with 4 hours reaction time, room temperature (27oC) with 4 hours reaction time, and reflux temperature (90oC) with 4, 8 and 16 hours reaction times. The reaction results were characterized using UV-Vis, IR, and GC-MS spectrophotometers. Reactions at ice temperature (0oC) and room temperature (27oC) with 4 hours reaction time of 4 hours do not produce organonitrogen compounds. While at reflux temperature (90oC) with 4 hours reaction time produces organonitrogen compounds, eugenyl acetamide. The reaction results at the reflux temperature (90oC) with 8, and 16 hours reaction times are organonitrogen compounds namely eugenyl acetamide compounds and isoquinolina compounds whose quantities increased with increasing reaction time. The reaction results at reflux temperature (90oC) with a reaction time of 16 hours was tested its toxicity level using the Brain Shrimp Lethality Test method to Artemia salina showed the value of LC50 was 16 ppm and classified as strong toxicity activity.
