Abstract
Penelitian mengenai perbandingan reaksi metilasi pada protein DMAP1 dan gen DMAP1 menggunakan katalis METTL10 bertujuan untuk melihat peluang serta pengaruh dari terjadinya reaksi metilasi antara protein DMAP1 dan gen DMAP1. Penelitian dilakukan dengan metode docking melalui AutoDock dan SwarmDock. Jarak antara ligan dengan reseptor dapat dilihat melalui Discovery Studio. Data yang didapatkan dari penelitian ini adalah energi ikatan (ΔG), konstanta inhibisi (Ki), entropi (ΔS), dan jarak interaksi ligan. Nilai ΔG yang didapatkan pada reaksi metilasi protein DMAP1 dan reaksi metilasi gen DMAP1 masing-masing sebesar -1,26 kcal/mol dan - 0,63 kcal/mol sehingga reaksi bersifat spontan. Konstanta inhibisi terkecil yang didapatkan pada reaksi metilasi protein DMAP1 dan reaksi metilasi gen DMAP1 masing-masing sebesar 7,73 mM dan 51,83 μM. Nilai ΔS yang dihasilkan pada reaksi metilasi protein DMAP1 dan reaksi metilasi gen DMAP1 membuktikan bahwa reaksi berlangsung secara kesetimbangan. Jarak yang dihasilkan pada interaksi AdoHcy dengan Fe4S4 tidak lebih dari 4Å, sedangkan jarak yang dihasilkan pada interaksi AdoMet dengan Fe4S4 lebih dari 4Å. Reaksi metilasi lebih cenderung terjadi pada tingkat protein. Reaksi metilasi pada protein DMAP1 diduga akan menyebabkan fungsi protein DMAP1 sebagai suppressor tumor menjadi tidak aktif.
English Abstract
Research on the comparison of methylation on DMAP1 protein and DMAP1 genes using METTL10 catalysts aims to see opportunity and the effects of the methylation reactions between DMAP1 protein and DMAP1 genes. The research was conducted by docking with AutoDock and SwarmDock. The distance between ligand and receptors can be viewed using Discovery Studio. Obtained data from this research are bond energy (G), inhibition constant (Ki), entropy (ΔS), and ligand interaction distance (R). The value of G in the DMAP1 protein methylation reaction and DMAP1 genes methylation reaction were respectively of -1.26 kcal/mol and -0.63 kcal/mol so the reaction was spontaneous. The smallest inhibition constants obtained in the DMAP1 protein methylation reaction and the DMAP1 gene methylation reactions were 7.73 mM and 51.83 μM respectively. The ΔS value generated in the methylated reaction of DMAP1 protein and the methylated reaction of DMAP1 gene proves that the reaction in equilibrium. The distance generated in the AdoHcy interaction with Fe4S4 is not more than 4Å, while the distance generated in the AdoMet interaction with Fe4S4 is more than 4Å. Methylation reactions are more likely to occur at the protein level. The methylation reaction in DMAP1 protein is suspected to cause the function of DMAP1 protein as tumor suppressor to be inactive.
