Abstract

Padi berperan sebagai pemenuh kebutuhan pokok karbohidrat bagi penduduk Indonesia. Oleh karena itu, produktivitas pertanian padi diharapkan mengalami peningkatan termasuk di wilayah Kabupaten Merauke, Provinsi Papua yang daya dukung tanah pertaniannya masih perlu diperbaiki. Tujuan penelitian adalah mengetahui potensi dan mengidentifikasi bakteri rhizosfer dari lahan padi di Kabupaten Merauke, Provinsi Papua sebagai penghasil IAA dan pelarut fosfat. Isolasi bakteri penghasil IAA dan pelarut fosfat masing-masing menggunakan media TSA yang mengandung triptofan 1 μg/mL dan Pikovskaya agar. Isolat dipilih berdasarkan karakter morfologi koloni dan isolat pembentuk zona bening sebagai karakter tambahan bakteri pelarut fosfat. Uji potensi bakteri penghasil IAA didapatkan tiga isolat (MI 10, MI 23 dan MI 25) dari 25 isolat dan uji potensi bakteri pelarut fosfat diperoleh empat isolat (MF 4, MF 14, MF 18 dan MF 21) dari 21 isolat. Bakteri yang berpotensi menghasilkan IAA tertinggi adalah isolat MI 25 sebesar 21,38 μg/mL pada waktu inkubasi 48 jam, sedangkan yang berpotensi melarutkan fosfat tertinggi adalah isolat MF 18 sebesar 14,44 μg/mL pada waktu inkubasi 96 jam. Isolat MI 25 dan MF 18 masing-masing memiliki similaritas tertinggi dengan Cedecea lapagei CIP 105857Τ sebesar 99% dan Burkholderia vietnamensis DSM 4587Τ sebesar 99%. Kedua bakteri tersebut berpotensi digunakan sebagai agen pupuk hayati, khususnya dalam meningkatkan produktivitas pertanian padi.

English Abstract

Rice plays an important role as main carbohydrate source in Indonesia. Therefore, productivity of paddy is expected to be increased, including in Merauke Regency of Papua Province, which is the soil bearing capacity is still required to be improved. This study was aimed to examine the potency and identify of isolated rhizobacteria from paddy field in Merauke Regency, Papua Province in producing indole-3-acetic acid (IAA) and in solubilizing phosphate. Isolation of rhizobacteria as IAA producer and phosphate solubilizer was conducted using TSA containing tryptophan 1 μg/mL and Pikovskaya agar, respectively. Isolates were firstly screened based on morphological characters and clear zone around the colonies for phosphate solubilizer was observed. There were three isolates (MI 10, MI 23 and MI 25) from 25 isolates that able to produce IAA and four isolates (MF 4, MF 14, MF 18 and MF 21) from 21 isolates that able to solubilize phosphate. The highest IAA was produced by MI 25 by 21.38 μg/mL at incubation time of 48 hours, while the best phosphate solubilizing bacteria was showed by isolate of MF 18 by 14.44 μg/mL at incubation time of 96 hours. MI 25 and MF 18 isolates were closest to Cedecea lapagei CIP 105857Τ with 99% of similarity and Burkholderia vietnamensis DSM 4587Τ with 99% of similarity, respectively. Both bacterial isolates are potentially to be utilized as biofertilizer specially for increasing productivity of paddy agriculture.
