Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh lama pretreatment alkali terhadap konsentrasi kolagen sisik ikan Gabus (C. striata) hasil ekstraksi dengan enzim bromelain dan profil protein kolagen tersebut. Pretreatment alkali dilakukan dengan perendaman sisik pada larutan NaOH 0,1 M selama 6 jam (P1) dan 72 jam (P2), kemudian dilanjutkan dengan dekalsifikasi menggunakan larutan EDTA 0,5 M selama 24 jam pada suhu 4 ºC. Selanjutnya, dilakukan ekstraksi kolagen dengan enzim bromelain (1 g sisik/ 10 mL bromelain). Hasil ekstraksi dari masing-masing perlakuan kemudian dibagi menjadi dua kelompok, yakni freeze dry dan non freeze dry. Ekstrak sisik ikan Gabus dari kelompok non freeze dry dipresipitasi terlebih dahulu, kemudian kedua kelompok sampel diukur konsentrasinya dengan metode Bradford pada panjang gelombang 630 nm. Selanjutnya dilakukan analisis SDS-PAGE menggunakan 8% separating gel untuk mengetahui profil protein yang didapatkan dari sisik ikan Gabus. Data konsentrasi protein kemudian dianalisis menggunakan uji Independent Sample T-test pada SPSS 22.0. Hasil penelitian ini menunjukkan bahwa ekstrak kolagen P1 memiliki konsentrasi lebih tinggi dibandingkan P2, baik pada sampel freeze dry maupun non freeze dry. Hasil separasi protein dengan SDSPAGE menunjukkan adanya perbedaan pola band protein yang muncul pada sampel P1 dan P2. Sampel 1 menunjukkan adanya komponen rantai α1 dan α2 serta subunit β dan γ, sedangkan pada sampel P2 tidak terdeteksi adanya rantai α. Keberadaaan komponen rantai α1 dan α2 pada hasil ekstraksi tersebut merupakan kolagen tipe I, dengan komposisi rantai α1 (125 KDa), dan α2 (152 KDa).

English Abstract

The aims of this study are to determine the effect of alkaline pretreatment on collagen concentration and protein profile of Snakehead (C. striata) fish scales collagen extracted with bromelain enzyme. Alkali pretreatment was performed by soaking the scales on 0.1 M NaOH solution for 6 hours (P1) and 72 hours (P2), followed by decalcification using 0.5 M EDTA solution for 24 hours at 4ºC. After that, the collagen extraction with bromelain enzyme (1 g of pretreated scales / 10 mL of bromelain) was performed. The extraction results from each treatment were then divided into freeze dry and non freeze dry group. The extract of Snakehead fish scales from the non freeze dry group was precipitated first, then the concentration of the two sample groups were measured in by Bradford method at 630 nm. Furthermore, SDS-PAGE analyzes with 8% separating gel to determine protein profile obtained from Snakehead fish scales was performed. Protein concentration data were further analysed using Independent Sample T-test in SPSS 22.0. The results of this study showed that P1 had higher collagen concentrations than P2, both in freeze dry and non freeze dry samples. The result of protein separation with SDS-PAGE showed the difference of protein band pattern that appeared on P1 and P2. The presence of α1 and α2 chain components as well as β and γ subunits were detected in P1, whereas in P2, no α chain is detected. The presence of the α1 and α2 chain components in the extract of Snakehead (C. striata) fish scale using bromelain enzyme showed that the collagen obtained was type collagen I, with α1 (125 KDa), and α2 (152 KDa) chain composition.
