Abstract

Esterifikasi terhadap komponen minyak sereh wangi dilakukan untuk menghasilkan tipe aroma yang berbeda dengan tipe aroma minyak sereh wangi awal. Esterifikasi komponen minyak sereh wangi dilakukan dengan membandingkan dua sumber asetil yaitu asam asetat glasial dan anhidrida asetat dengan asam fosfat sebagai katalis melalui proses refluks 3 jam dan 7 jam. Produk esterifikasi dikarakterisasi menggunakan GC-MS, sedangkan profil aroma diuji secara organoleptik menggunakan panelist dan didukung oleh pendekatan simulasi dockng molekuler (in silico). Esterifikasi minyak sereh wangi dengan asam asetat selama 7 jam refluk menghasilkan 5,57% sitronelil asetat dan 4,98% geranil asetat, sedangkan jika dengan anhidrida asetat selama 3 jam refluk menghasilkan 35.15% sitronelil asetat dan 36.55% geranil asetat. Esterifikasi fraksi minyak sereh yang mengandung komponen dominan geraniol dan sitronelol menggunakan asam asetat selama 7 jam refluk menghasilkan 33.37% sitronelil asetat dan 4.104% geranil asetat. Uji perbedaan kesukaan aroma menunjukkan bahwa 65% panelist lebih menyukai aroma hasil esterifikasi daripada aroma bahan dasarnya, disebakan aroma produk esterifikasi lebih floral, fruity dengan latar rose. Interaksi molekul ester dengan reseptor OR1A1 (yang merespon aroma sweet, floral, fruity,rose) dan OR1A2 (yang merespon sweet, floral,rose) menunjukkan sisi pengikatan residu asam amino yang hampir sama. Adapun afintasnya lebih kuat pada reseptor dengan OR1A2. Berdasarkan penilaian Kd, afinitas ester sitronelil asetat dan geranil asetat lebih kuat daripada senyawa sitronelol dan geraniol.

English Abstract

Esterification of citronella oil component was carried out to produce different types of aroma relative to the aroma of originearly citronella oil. Esterification of citronella oil component was done by comparing two sources of acetyl i.e glacial acetic acid and acetic anhydride with phosphoric acid as catalyst through reflux process for 3 hours and 7 hours. The esterification product was characterized using GC-MS, while the aroma profile was tested organoleptically using a panelist and supported by a molecular docking (in silico) simulation approach. Esterification of citronella oil with acetic acid for 7 hours of reflux yielded 5.57% citronellyl acetate and 4.98% geranil acetate, whereas if with acetic anhydride for 3 hours reflux yield 35.15% citronellyl acetate and 36.55% geranil acetate. Esterification of distilate fraction of citronella oil in which containing the dominant components of geraniol and citronellol using acetic acid for 7 hours of reflux yielded 33.37% citronellyl acetate and 4.104% geranil acetate. The difference test of aroma preferences shows that 65% panelist prefer the smell of esterification result rather than the aroma of the basic material due to tipe of aroma more floral, fruity with rose background. The interaction of ester molecules with OR1A1 receptors (which responds to sweet, floral, fruity, rose) and OR1A2 (which responds to sweet, floral, rose) indicates that the interaction was at the same side of the amino acid residue binding. The affinity of Ligand-Receptor binding was more strongly on the receptor with OR1A2. Based on scoring of the Kd, the affinity of the citronellyl acetate and the geranyl acetate more higher than citronellol and geraniol.
