Abstract
Senyawa turunan benzokuinon 2-(5-bromopentil)-3,5,6-trimetil-1,4- benzokuinon telah disintesis melalui reaksi oksidasi dan reaksi alkilasi. Reaksi oksidasi dari senyawa 2,3,5-trimetil-1,4-hidrokuinon menghasilkan senyawa 2,3,5-trimetil-1,4-benzokuinon. Reaksi alkilasi dengan asam bromoheksanoat menghasilkan senyawa 2-(5- bromopentil)-3,5,6-trimetil-1,4-benzokuinon (TMBQ5). Produk TMBQ5 diperoleh dengan yield sebesar 32,59%. Sintesis dilakukan untuk memperoleh senyawa dengan lipofilisitas lebih tinggi dibandingkan timokuinon, sehingga dapat menembus membran lipid bilayer lebih baik. Senyawa hasil sintesis diuji koefisien partisinya dalam sistem oktanol-air dan dibandingkan dengan timokuinon. Senyawa TMBQ5 memiliki nilai log P 2,23 sedangkan timokuinon 2,50. Senyawa TMBQ5 diprediksi memiliki kemampuan menembus membran lipid bilayer yang setara dengan timokuinon.
English Abstract
Synthesis of 2-(5-bromopentyl)-3,5,6-trimethyl-1,4-benzoquinone was carried out by oxidation and alkylation reaction. The oxidation reaction was performed by reacted 2,3,5-trimethyl-1,4-hydroquinone to get 2,3,5-trimethyl-1,4-benzoquinone. The alkylation reaction using bromohexanoic acid gave 2-(5-bromopenthyl)-3,5,6-trimethyl- 1,4-benzoquinone (TMBQ5). The yield of TMBQ5 was 32.59%. Synthesis was performed to obtain compounds with higher liphopilicity compared with thymoquinone, so it can pass through lipid bilayer membrane better. TMBQ5 was tested their partition coefficient in an octanol-water system and compared with thymoquinone. TMBQ5 gave log P 2.23 and tymoquinone is 2.50. By this result, TMBQ5 was predicted to have similar ability to pass through lipid bilayer membrane than thymoquinone.
