Abstract
Hipertensi merupakan suatu penyakit kelainan tekanan darah, dimana tekanan sistolik dan diastolik dalam keadaan diatas normal yaitu >140 mmHg dan >90 mmHg. Salah satu penyebab hipertensi adalah tingginya kadar radikal bebas dalam tubuh yang diperoleh melalui jalur RAAS atau jalur inflamasi. Pembuatan kondisi hipertensi menggunakan induksi Deoxycorticosterone Acetate (DOCA)-Salt dapat menyebabkan peningkatan stres oksidatif dan inflamasi pada jantung sehingga meningkatkan tekanan darah. Salah satu obat herbal yang mengandung antioksidan untuk menetralkan radikal bebas adalah ekstrak ubi jalar ungu (Ipomoea batatas L. var Ayamurasaki). Penelitian ini bertujuan untuk mengetahui potensi ekstrak ubi jalar ungu dalam menurunkan kadar Malondialdehyde (MDA) dan memperbaiki jantung tikus model hipertensi induksi DOCA-Salt. Penelitian dibagi dalam empat kelompok perlakuan yaitu kontrol negatif (normal); kontrol positif (hipertensi); terapi ekstrak ubi jalar ungu 200 mg/kgBB; terapi 2 ekstrak ubi jalar ungu 400 mg/kgBB. Berdasarkan hasil, diperoleh kadar MDA untuk masingmasing perlakuan yaitu 0,184 ± 0,0067 µg/mL; 0,672 ± 0,0084 µg/mL; 0,402 ± 0,0050 µg/mL; 0,263 ± 0,0064 µg/mL. Terapi antosianin dalam ekstrak ubi jalar ungu dapat menurunkan kadar MDA hingga 40,11% untuk dosis 200 mg/kgBB dan 60,18% untuk dosis 400 mg/kgBB. Hasil statistik untuk kadar MDA memiliki nilai signifikansi p<0,01 dan memiliki perbedaan yang sangat nyata antar perlakuan. Terapi ekstrak ubi jalar ungu memberikan efek pada perbaikan histopatologi jantung pada struktur myosit dan myofibril. Berdasarkan hasil penelitian, dapat disimpulkan bahwa terapi ekstrak ubi jalar ungu dapat menurunkan kadar MDA dan memperbaiki gambaran histopatologi jantung tikus hipertensi induksi DOCA-Salt.
English Abstract
Hypertension is a blood pressure disorder, where systolic and diastolic blood pressure is over 140 mmHg and 90 mmHg respectively. Based on the previous research, it is known that one of the cause of hypertension is high level of free radicals that produced through RAAS or inflammation. Hypertension induced by Deoxycorticosterone Acetate (DOCA)-Salt may leads to increasing of oxidative stress and inflammantory effect of the heart thus increasing blood pressure. One of herbal remedies that contains high level of antioxidants to neutralize free radicals is purple sweet potato (Ipomoea batatas L. var Ayamurasaki) extract. The aim of this research was to study the effect of purple sweet potato extract in lowering Malondialdehyde (MDA) level and repairing heart of rats (Rattus norvegicus). This study was divided into four treatment groups; negative control (normal); positive control (hypertension); therapy 1 (purple sweet potato extract (200 mg/kgBB); therapy 2 (purple sweet potato 400 mg/kgBB). Based on the analysis, MDA levels obtained for each treatment; 0,184 ± 0,0067 µg/mL; 0,672 ± 0,0084 µg/mL; 0,402 ± 0,0050 µg/mL; 0,263 ± 0,0064 µg/mL, which indicated therapy using purple sweet potato extract can lower MDA level up to 74,80% in dose of 200 mg/kgBB extract, and 84,44% in dose of 400 mg/kgBB. The statistical results for MDA level has shown significance value of p<0,01 and give a very significance different between treatments. The purple sweet potato extract also showed repairing in histopathology of the heart especially myosit and myofibril structres. Based of the results of the study, it can be concluded that purple sweet potato extract can be used for lowering MDA level and repairing the histopathology of heart.
