Abstract
Diabetes Mellitus dapat menimbulkan komplikasi makrovaskuler dan mikrovaskuler. Salah satu contoh komplikasi mikrovaskuler yaitu nefropati diabetik. Dalam patogenesis nefropati diabetik, stres oksidatif mengaktifkan jalur patogen lainnya, yang akhirnya menyebabkan kerusakan fungsi ginjal yang ditandai dengan peningkatan Glomerular Filtration Rate (GFR). Dengan demikian, penghambatan stres oksidatif merupakan titik fokus untuk beberapa sinergi terapeutik. Asam rosmarinat (AR) dan telmisartan dapat memberikan harapan pada tatalaksana nefropati diabetik. Cystatin C dianggap sebagai penanda fungsi ginjal yang baik karena cystatin C difiltrasi oleh glomerulus, diserap kembali dan dikatabolisme sepenuhnya oleh tubulus proksimal, serta berkorelasi dengan GFR. Tujuan penelitian ini adalah untuk mengetahui perbandingan efek asam rosmarinat dan telmisartan terhadap kadar cystatin C serum pada tikus (Rattus norvegicus) galur wistar model diabetes melitus dengan desain penelitian true experimental laboratory randomized posttest only controlled group design. Berdasarkan hasil penelitian, pada tikus dengan terapi asam rosmarinat mengalami penurunan konsentrasi cystatin C sebesar 28,80%, sedangkan tikus dengan terapi telmisartan mengalami penurunan sebesar 19,63% dibandingkan tikus kontrol (diabetes). Berdasarkan hasil analisis Uji OneWay ANOVA, terdapat perbedaan bermakna diantara kelompok perlakuan (p < 0,05). Jadi dapat disimpulkan bahwa pemberian asam rosmarinat lebih baik dibandingkan dengan telmisartan dalam mencegah peningkatan kadar cystatin C serum pada tikus (Rattus norvegicus) galur wistar model diabetes melitus
English Abstract
Diabetes Mellitus can cause macrovascular and microvascular complications. One example of microvascular complications is diabetic nephropathy. In the pathogenesis of diabetic nephropathy, oxidative stress activates other pathogenic pathways, which eventually causes damage to kidney function that is marked with an increased of Glomerular Filter Rate (GFR). Thus, inhibition of oxidative stress is the focal point for some therapeutic synergies. Rosmarinic acid (RA) and telmisartan can give hope in diabetic nephropathy treatment. Cystatin C is considered as a marker in renal dysfunction because the cystatin C free filtrated by the glomerulus, reabsorbed and catabolized by proximal tubules, and highly correlated with GFR. The purpose of this research was to know the comparison between effect of rosmarinic acid and cystatin C serum levels against telmisartan on rats (Rattus norvegicus) strain wistar model of diabetes mellitus. Design of this research was randomized post test only controlled group design. Based on the research, the results showed in mice with rosmarinic acid therapy there was decreased the concentration of cystatin C level aproximately to 28,80%, while mice with telmisartan therapy decreased by 19,63%. Based on the results of one-way ANOVA test, there were significance different among groups (p<0,05). So it can be concluded that administration of rosmarinic acid is better than telmisartan in preventing elevated serum cystatin C levels in rats (Rattus norvegicus) wistar strains in the diabetes mellitus model
