Abstract
Diabetes melitus merupakan penyakit metabolik yang sering dijumpai saat ini dan diketahui dapat menyebabkan berbagai komplikasi salah satunya yaitu kerusakan sistem saraf motorik. Genistein diketahui memiliki manfaat untuk pengobatan penyakit gula darah tinggi dan mencegah komplikasinya. Penelitian ini bertujuan untuk membuktikan efek pemberian genistein terhadap penghambatan penurunan respon taktil pada zebrafish yang diinduksi glukosa tinggi. Studi eksperimental menggunakan laboratorium post test-only control group design dilakukan terhadap hewan coba zebrafish. Sampel dipilih dengan cara simple random sampling lalu dibagi menjadi enam kelompok, yaitu kelompok kontrol negatif (embrio medium), kontrol positif (glukosa 3%), genistein 2 μm, glukosa 3% + genistein 0,5 μm, glukosa 3% + genistein 1 μm, dan glukosa 3% + genistein 2 μm. Variabel yang diukur adalah respon taktil pada embrio zebrafish usia 72 hpf. Hasil penelitian menunjukkan bahwa hasil respon taktil embrio zebrafish usia 72 hpf pada kelompok kontrol positif (glukosa 3%) lebih rendah bila dibandingkan dengan kelompok genistein dosis 0,5 μm, 1 μm, maupun 2 μm. Berdasarkan penelitian ini, dapat disimpulkan bahwa genistein memiliki efek untuk menghambat penurunan respon taktil embrio zebrafish usia 72 hpf akibat paparan glukosa tinggi.
English Abstract
Diabetes mellitus is the common disease in this era. This disease has known as disease that has many complications such as defect of motor neuron system. Genistein has known as substance that can be used to therapy for this disease and its complication. This research is aimed to prove genistein effect to improve tactile stimulation in zebrafish with hyperglicemia. Experimental study using post test-only control group design was conducted onto zebrafish. The samples selected by simple random sampling to be divided into six groups, that is negative control (embrio medium), positive control (glukosa 3%), genistein 2 μm, glucose 3% + genistein 0,5 μm, glucose 3% + genistein 1 μm, and glucose 3% + genistein 2 μm. The variable measured was tactile stimulation in embrio zebrafish 72 hpf. Result of this research indicated that tactile stimulation in positive group (glucose 3%) has lower value than value in group with genistein 0,5 μm, 1 μm, and 2 μm. The conclusion is genistein has effect to inhibit decrease of tactile stimulation in embrio zebrafish 72 hpf after the exposure of high glucose.
