Abstract
Staphylococcus aureus merupakan agen penyebab infeksi piogenik pada manusia yang paling sering terjadi. Antibiotik golongan beta laktam merupakan lini pertama untuk mengatasi infeksi bakteri Staphylococcus aureus, namun pada kondisi rumah sakit, strain Staphylococcus aureus yang ditemukan kebanyakan mengalami resistensi. Untuk itu, diperlukan suatu terapi yang berasal dari tanaman, seperti rumput teki (Cyperus rotundus). Tujuan penelitian ini adalah untuk mengetahui efek dari pemberian ekstrak kasar rimpang rumput teki (Cyperus rotundus) terhadap Staphylococcus aureus secara in vitro. Metode yang digunakan dalam penelitian adalah eksperimental laboratories secara in vitro menggunakan dilusi tabung dan Microwave Assisted Extraction. Konsentrasi ekstrak rimpang rumput teki (Cyperus rotundus) yang digunakan dalam penelitian ini adalah 0%, 5%, 10%, 15%, 20%, 25% dan 30%. Kadar Hambat Minimal (KHM) yang ditemukan ada pada konsentrasi 10%, sedangkan Kadar Bunuh Minimal (KBM) yang ditemukan ada pada konsentrasi 20%. Hasil dari uji normalitas Shapiro-Wilk didapatkan nilai sig p=0,416 (p>0,05) dan uji homogenitas Levene didapatkan nilai sig p=0,139 (p>0,05). Untuk uji komparatif One-way Annova, didapatkan nilai sig p=0,000 (p<0,05). Hasil dari uji regresi linier sederhana adalah R Square= 0,682 yang berarti kemampuan ekstrak rimpang rumput teki (Cyperus rotundus) dalam menurunkan jumlah koloni bakteri Staphylococcus aureus adalah sebesar 68%. Uji korelasi Pearson memberikan hasil nilai r=-0,826 yang menunjukkan semakin tinggi konsentrasi ekstrak rimpang rumput teki (Cyperus rotundus) maka jumlah koloni bakteri Staphylococcus aureus akan semakin rendah. Berdasarkan penelitian tersebut, dapat disimpulkan bahwa ekstrak rimpang rumput teki (Cyperus rotundus) dapat berperan sebagai antibakteri terhadap bakteri Staphylococcus aureus.
English Abstract
Staphylococcus aureus is the most common agent that causes pyogenic infections in humans. Beta lactam antibiotics are the first line therapy for the infections caused by Staphylococcus aureus, but in hospital conditions, the strains of Staphylococcus aureus mostly have antibiotics resistance. Thus, a new plantbased therapy is needed, using plants such as nut grass (Cyperus rotundus). The purpose of this study was to determine the effect of the of nut grass (Cyperus rotundus) crude extract to Staphylococcus aureus through in vitro application. The method used in the study was experimental laboratories using tube dilution method and Microwave Assisted Extraction. The concentration of the nut grass crude extract (Cyperus rotundus) used for this study was 0%, 5%, 10%, 15%, 20%, 25% and 30%. The Minimum Inhibitory Level (MIC) was found at a concentration of 10%, while the Minimum Bactericidal Concentration (MBC) was found at a concentration of 20%. The results of the Shapiro-Wilk normality test obtained a sig value p = 0.416 (p> 0.05) and the Levene homogeneity test obtained sig p = 0.139 (p> 0.05). For the One-way Annova comparative test, sig p = 0,000 (p <0.05) was obtained. The results of a simple linear regression test are R Square = 0.682 which means the ability of the crude extract of nut grass (Cyperus rotundus) in reducing the number of colonies of Staphylococcus aureus bacteria is 68%. Pearson correlation test gave the results of the value r = -0.826 which showed that the higher the concentration of the extract, will make the number of Staphylococcus aureus bacteria colonies goes lower. Based on these studies, it can be concluded that the crude extract of the rhizome of nut grass (Cyperus rotundus) can act as an antibacterial against the Staphylococcus aureus
