Abstract
Luka insisi adalah salah satu luka terbuka dimana terjadi rusaknya struktur dan fungsi anatomis kulit normal akibat proses eskternal oleh instrument tajam dan menimbulkan inflamasi akut dan menyebabkan jumlah sel radang dan ekspresi sitokin proinflamasi IL-1 mengalami kenaikan. Minyak biji kelor (Moringa olifera) mengandung bahan aktif flavanoid berpotensi sebagai antiinflamasi sehingga proses penyembuhan luka dapat berjalan lebih cepat. Penelitian bertujuan untuk mengetahui pengaruh pemberian minyak biji kelor (Moringa olifera) terhadap jumlah sel radang dan ekspresi IL-1 pada area luka. Penelitian dilaksanakan secara eksperimental, digunakan hewan coba tikus putih (Rattus norvegicus) jantan galur wistar 200 gr. Penelitian menggunakan Rancangan Acak Lengkap (RAL) dibagi dalam 5 kelompok perlakuan yaitu kelompok kontrol negatif (KN) tanpa perlakuan, kelompok kontrol positif (KP) diinsisi tanpa terapi, kelompok P1 diinsisi dan diterapi minyak biji kelor 50%, P2 diinsisi dan diterapi minyak biji kelor 75%, P3 diinsisi dan diterapi minyak biji kelor 100%. Pemberian terapi dilakukan dua kali sehari secara topikal sebanyak 50 μL selama 7 hari. Parameter sel radang diamati menggunakan pewarnaan HE (Hematoxyline eosin) dan IL-1 menggunakan IHK (imunohistokimia). Data kuantitatif dianalisa statistik one-way ANOVA diuji lanjutan Tukey α= 0,05. Hasil penelitian menunjukan kelompok terapi P1, P2, P3 menurunkan jumlah sel radang dan ekspresi IL-1. Pada perhitungan jumlah sel radang menunjukan hasil KN (4,00 ± 1,25), KP (38.25 ± 1,70), P1 (21,50 ± 1,29), P2 (10,75 ± 0,95), P3 (16,75 ± 0,95) sel sedangkan ekspresi IL-1 menunjukkan presentase KN (05,00 ± 0,81), KP (25,25 ± 01,70), P1 (21,50 ± 1,29), P2 (10,75 ± 0,95), P3 (16,75 ± 0,95). Kesimpulan penelitian ini adalah kelompok terapi P2 memiliki efek terapi yang paling baik untuk terapi luka insisi dengan menurunkan jumlah sel radang dan presentasi ekspresi IL-1.
English Abstract
Incision wound is one of the open sores where there is damage to the structure and anatomical function of normal skin due to external processes by the sharp instrument and cause acute inflammation therefore increasing the number of inflammatory cells and expression of IL-1 proinflammatory cytokines. Moringa seed oil (Moringa olifera) contains flavanoid active ingredients that have the potential to be anti-inflammatory so that the wound healing process can run faster. The aim of this study was to determine the effect of Moringa olifera seed oil on the number of inflammatory cells and IL-1 expression in the wound area. The experiment was carried out experimentally, using animal experiments on male white rats (Rattus norvegicus) wistar strain 200 gr. The study used Completely Randomized Design divided into 5 groups: negative control group (KN) without treatment, positive control group (KP) incised without therapy, P1 incised and treated with 50% moringa seed oil, P2 incised and treated with 75% moringa seed oil, P3 incised and treated with 100% moringa seed oil. Topical therapy was administered twice daily for 50 μL for 7 days. Inflammatory cell parameters were observed using HE staining (Hematoxyline eosin) and IL-1 using immunohistochemistry. Quantitative data analyzed statistically into one-way ANOVA tested further with Tukey α = 0.05. The results showed that the P1, P2, P3 therapy groups reduced the number of inflammatory cells and IL-1 expression. In the calculation of the number of inflammatory cells showed the results of KN (4,00 ± 1,25), KP (38.25 ± 1,70), P1 (21.50 ± 1.29), P2 (10.75 ± 0.95), P3 (16.75 ± 0.95) cells while the expression IL-1 showed a percentage of KN (05,00 ± 0,81), KP (25,25 ± 01,70), P1 (21 , 50 ± 1.29), P2 (10.75 ± 0.95), P3 (16.75 ± 0.95). The conclusion of this study was that the P2 therapy group had the best therapeutic effect for incision wound therapy by reducing the number of inflammatory cells and the presentation of IL-1 expression.
