Abstract

Malondialdehida (MDA) merupakan senyawa yang dihasilkan dari peroksidasi lipid akibat tingginya radikal bebas dalam tubuh. Aflatoksin merupakan salah satu metabolit sekunder yang diproduksi oleh spesies kapang dari genus Aspergillus, yaitu Aspergillus flavus, Aspergillus parasiticus, dan Aspergillus nomius yang dapat meningkatkan radikal bebas yang dapat menyebabkan kematian sel hepar. Kemampuan bakteri asam laktat dalam mengikat aflatoksin akan mencegah nekrosis sel hepar dengan cara menghambat absorbsi aflatoksin, baik secara in vitro maupun in vivo. Penelitian ini bertujuan efek pemberian Lactobacillus plantarum terhadap kadar MDA serta hispotapatologi hepar tikus putih (Rattus norvegicus) yang diinduksi aflatoksin dari Aspergillus flavus. Penelitian ini bersifat eksperimental dengan desain post test only control group menggunakan Rancangan Acak Lengkap (RAL). Hewan model menggunakan tikus putih (Rattus norvegicus) strain Wistar jantan dengan umur 8 – 10 minggu dan berat badan 150 – 250 g yang dibagi dalam lima kelompok perlakuan. Perbandingan kelompok negatif dengan positif yaitu diberi aflatoksin dengan dosis 7 μg/kg BB secara peroral selama 21 hari, serta perlakuan 1, 2, dan 3 yang disuplementasi BAL dosis 1 ml dengan konsentrasi masing-masing 1x104, 1x106, dan 1x108 CFU/ml secara peroral selama 21 hari serta induksi aflatoksin secara peroral mulai hari ke 8 – 28. Kadar kerusakan diukur secara kuantitatif menggunakan MDA serta dianalisis statistika dengan uji ragam One-Way ANOVA, α = 5%. Sementara hasil pengamatan histopatologi hepar berdasarkan gambaran kerusakan sel hepar yang dianalisa secara deskriptif. Hasil didapatkan pada kelompok perlakuan 2 dengan konsentrasi BAL 1x106 CFU/ml memberi pengaruh terbaik terhadap pencegahan peningkatan kadar MDA dan mencegah kerusakan hepar. Kesimpulan dari penelitian ini adalah BAL lactobacillus plantarum dapat mencegah aflatoksikosis dengan mencegah kenaikan kadar MDA dan kerusakan gambaran histopatologi hepar.

English Abstract

Malondialdehyde (MDA) is a compound which resulted from lipid peroxidation due to a high amount of free radicals in the body. Aflatoxin is one of the secondary metabolites produced by species of mold from genus of Aspergillus, which are Aspergillus flavus, Aspergillus parasiticus and Aspergillus nomius that could increase the amount free radicals and lead to the death of liver cells. Lactic acid bacteria abilities in binding the aflatoxin would prevent liver cells necrosis by inhibiting aflatoxin absorption, by in vivo as well as in vitro. The purpose of this study was to find out the effect of Lactobacillus plantarum on MDA levels and liver histopathological appearance of white rats (Rattus norvegicus) induced by aflatoxin from Aspergillus flavus. This study was experimental using Completely Randomized Design (RAL) and drafted by post-test only control group design. Male Wistar rats age between 8 – 10 weeks and weigh 150 – 250 gr were used as the model animals and were divided into five groups. The comparison between the positive group and the negative group was given aflatoxin with dose 7 μg/kg BW orally for 21 days, and also the treatment 1, 2 and 3 which supplemented by BAL (Lactic acid bacteria) with dose of 1 ml with each concentrations of 1x104, 1x106, dan 1x108 CFU/ml orally for 21 days and the induction of aflatoxin orally was started from day 8 until day 28. Damage levels were measured quantitatively using MDA and have been statistically analyzed with One-Way ANOVA method, α = 5%. Observation result of liver histopathological image based on the appearance of liver damage that occurred was analyzed descriptively. The result found in the 2nd group with the concentration of 1x106 CFU/ml BAL has shown the best effect on preventing the increase levels of MDA and preventing liver damage. The conclusion from this study was lactic acid bacteria (LAB) of Lactobacillus plantarum could prevent aflatoxicosis by preventing the increase levels of MDA and liver histopathological damage appearance.
