Abstract

Dewasa ini masyarakat Indonesia sadar akan pentingnya konsumsi protein hewani, salah satunya yaitu kambing. Namun, produksi nasional daging kambing menunjukkan penurunan. Permasalahan ini dapat diatasi dengan menerapkan bioteknologi reproduksi seperti inseminasi buatan dan fertilisasi in vitro. Dalam proses fertilisasi in vitro terdapat beberapa hambatan yang dapat menurunkan keberhasilan produksi embrio, salah satunya yaitu polispermia. Prevalensi polispermi pada domba besarannya sampai 20%. Penelitian sebelumnya menunjukkan bahwa paparan gamet oleh sel epitel oviduk dan sekresi oviduk dapat mengurangi polispermi. Melihat potensi tersebut maka dalam penelitian ini bertujuan untuk mengetahui pengaruh suplementasi Ephitelia Cell Oviductin Glycoprotein pada media kapasitasi terhadap angka fertilisasi secara in vitro. Dalam penelitian ini spermatozoa yang digunakan disuplementasi dengan Ephitelia Cell Oviductin Glycoprotein sebelum dikapasitasi. Adapun media pengencer spermatozoa dan fertilisasi adalah media fertilisasi. Fertilisasi in vitro dilakukan selama 2 jam menggunakan media media fertilisasi dan diinkubasi selama 24 jam sehingga didapatkan zigot dua sel. Proses inkubasi dilakukan dalam inkubator Oxoid 4% CO2 dengan temperatur 37,3oC. Proses suplementasi dibagi menjadi 3 kelompok perlakuan, yaitu kelompok K sebagai kontrol (media Kapasitasi tidak ditambahkan ECOG) ; kelompok 1 (media Kapasitasi ditambahkan ECOG sebanyak 5 μl/ml) ; kelompok 2 (media Kapasitasi ditambahkan ECOG sebanyak 10 μl/ml). Hasil fertilisasi in vitro dianalisis dengan menghitung oosit yang tidak berbentuk zigot dan oosit yang menunjukkan adanya polar body II. Penelitian dilaksanakan secara eksperimen dengan menggunakan Rancangan Acak Lengkap. Data dianalisa secara statistik dengan uji F, dan bila signifikan dilanjutkan uji BNT. Hasil penelitian menunjukkan suplementasi ECOG 10 μl/ml memberikan angka fertilisasi tertinggi (90%) dan angka tidak terfertilisasi terendah (10%). Kesimpulan dari penelitian ini adalah pemberian suplementasi Ephitelia Cell Oviductin Glycoprotein pada media kapasitasi dapat meningkatkan angka fertilisasi secara in vitro.

English Abstract

Today the Indonesian people are aware of the importance of the consumption of animal protein, one of which is the goat. However, the national production of goat meat showed a decline. This problem can be solved by applying reproductive biotechnology such as artificial insemination and in vitro fertilization. In vitro fertilization process there are several obstacles that can decrease the success of embryo production, one of them is polispermia. Polispermi prevalence in sheep amount up to 20%. Previous research has shown that gamete exposure by oviduct epithelial cells and oviduct secretion may reduce polyspermi. Seeing this potential then in this study aims to determine the effect of supplementation Ephitelia Cell Oviductin Glycoprotein on the media of capacitation to fertilization rate in vitro. In this study the spermatozoa used was supplemented with Ephitelia Cell Oviductin Glycoprotein before it was captured. The spermatozoa sperm medium and fertilization is the fertilization medium. In vitro fertilization was performed for 2 hours using fertilization media medium and incubated for 24 hours to obtain a two-cell zygote. The incubation process is carried out in a 4% CO 2 Oxoid incubator with a temperature of 37.3 ° C. The supplementation process was divided into 3 treatment groups, namely group K as control (Capacitation media not added ECOG); group 1 (Capacity added ECOG medium 5 μl / ml); group 2 (Capacity added ECOG medium 10 μl / ml). The results of in vitro fertilization were analyzed by counting oocytes not in the form of zygotes and oocytes showing polar body II. The experiment was conducted experimentally using Completely Randomized Design. Data were analyzed statistically with F test, and if significant continued BNT test. The results showed that ECOG 10 μl / ml supplementation gave the highest fertilization rate (90%) and the lowest non-fertilization rate (10%). The conclusion of this research is supplementation of Ephitelia Cell Oviductin Glycoprotein on capacitation media can increase fertilization rate in vitro.
