Abstract
Hiperkolesterolemia merupakan keadaan meningkatnya kadar kolesterol di dalam darah yang melebihi kadar normal. Hewan hiperkolesterolemia akan mengalami peningkatan kadar trigliserida (TG). Angkak merupakan hasil fermentasi beras, dan mempunyai kandungan bioaktif seperti monakolin K (Lovastatin), dihidromonakolin, asam lemak tak jenuh yang berupa asam oleat, asam linoleat, asam linolenat, serta vitamin B kompleks yang berfungsi untuk pengelolaan kolesterol darah, diabetes, tekanan darah dan obesitas. Lovastatin yang terdapat pada angkak mampu menghambat HMG-CoA reduktase (Hydroxy-methyl-glutaryl Coenzyme A), yaitu enzim yang mengontrol jalur biosintesis kolesterol di dalam hati. Penelitian ini bertujuan untuk mengetahui terapi pemberian angkak terhadap induksi pakan diet hiperkolesterol yang diamati dari kadar trigliserida dan gambaran histopatologi hepar. Penelitian ini dilakukan bersifat eksperimental menggunakan Rancangan Acak Lengkap (RAL). Tikus putih (Rattus norvegicus) dibagi menjadi 5 kelompok, yaitu kelompok negatif, kelompok positif, kelompok dengan dosis angkak 0,5 g/ekor/hari; 1 g/ekor/hari; dan 1,5 g/ekor/hari. Pengujian kadar trigliserida menggunakan metode GPO-PAP. Analisa data trigliserida dengan one way ANOVA menggunakan SPSS-16. Pengamatan histologi hepar dilakukan secara deskriptif. Hasil penelitian menunjukkan bahwa terapi angkak dapat menurunkan kadar trigliserida secara signifikan (P<0,05). Pengamatan histopatologi menunjukkan bahwa angkak dapat menurunkan perlemakan dan memperbaiki gambaran histopatologi hepar. Dosis 1,5 g/ekor/hari menunjukkan dosis terbaik dalam menurunkan kadar trigliserida sebesar 15.44% dan dapat mengurangi perlemakan di hepar. Dapat disimpulkan bahwa angkak dapat menurunkan kadar trigliserida dan memperbaiki gambaran histopatologi hepar tikus model hiperkolesterolemia.
English Abstract
Hypercholesterolemia is a condition which the blood cholesterol level skyrocketed beyond the normal threshold. Animal with hypercholesterolemia will followed by the increase of triglyceride (TG) level. Angkak is the product of fermented rice that contains the bioactive substances, e.g. monacolin K (Lovastatin), dihydromonacolin, unsaturated fatty acid (oleic acid, linoleic acid, linolenic acid), and vitamin B complex which contributes to maintain blood cholesterol level, diabetes, blood pressure, and obesity. Lovastatin of angkak was known to inhibit HMG-CoA reductase (Hydroxy-methyl-glutaryl Coenzyme A) which is an enzime that controlled the hepatic cholesterol biosynthesis. This research was conducted to understand the therapeutic effect of angkak towards TG level and hepatic histological assessment of hipercholesterol diet animal model. This experimental research used Completely Randomized Design. White rats (Rattus norvegicus) was divided into 5 groups, which is negative group, positive group, daily 0,5g angkak/individual group, daily 1g angkak/individual group, and daily 1,5g angkak/individual group. The TG level was measured by GPO-PAP method and was analyzed with one-way ANOVA on SPSS-16. The hepatic histological assessment was carried out descriptively. The result revealed that angkak therapy reduced the TG level significantly (P<0,05). The observation of hepatic histology showed that angkak reduced tissue fat and mend the hepatic condition. The best effect was seen on daily 1,5 g angkak teraphy which reduced the TG level by 15.44% and reduced fatty condition in hepar. In conclusion, angkak teraphy have a possible use to supress trygliceride level and repair the hepatic histological condition on hypercholesterolemia rat model.
