Abstract

Osteoporosis terjadi karena adanya perubahan komposisi mineral tulang sehingga densitas masa tulang mengalami pengurangan dan menjadi rapuh sehingga berisiko terjadi patah tulang. Osteoporosis merupakan penyakit degeneratif yang paling sering menyerang wanita yang telah mengalami menopause, karena pada wanita menopause terjadi penurunan dari kadar estrogen. Penurunan kadar estrogen akan memacu aktivitas remodeling tulang yang tidak seimbang karena osteoblas tidak dapat mengimbangi kerja osteoklas. Pada penelitian sebelumnya, diketahui bahwa nikotin dapat meningkatkan aktivitas osteoblas in vitro, sehingga nikotin berpotensi sebagai terapi alternatif pada osteoporosis. Penelitian ini bertujuan untuk mengetahui pengaruh nikotin terhadap serum kalsitonin pada tikus model osteoporosis. Penelitian ini menggunakan tikus model osteoporosis dengan cara ovariektomi yang diberi nikotin dengan dosis 0.25 mg/KgBB, 0.5 mg/KgBB dan 0.75 mg/KgBB selama tujuh minggu pasca ovariektomi. Hasil penelitian menunjukkan bahwa ada perbedaan kadar kalsitonin pada perlakuan dosis (ANOVA, p=0.000), namun tidak ada hubungan antara dosis dan kadar kalsitonin (Spearman p=0.083, r=-0.353). Namun demikian, secara deskriptif terlihat kadar kalsitonin menurun seiring meningkatnya pemberian dosis, hasil yang tidak signifikan ini kemungkinan disebabkan karena prosedur ovariektomi sangat invasif sehingga tikus rawan terkena infeksi dan banyak tikus yang mati paska pembedahan. Kesimpulan penelitian ini bahwa pemberian nikotin cenderung meningkatkan kadar serum kalsitonin, yang dapat dilihat dari adanya peningkatan rata-rata kalsitonin seiring meningkatnya dosis pemberian.

English Abstract

Osteoporosis occurs as changes in bone mineral composition which causes the bone mass density is reduced and becomes brittle, so it would create a risk of fractures. Osteoporosis is also a degenerative disease that mostly attacks women who have experienced on the menopause stage. Since in the menopausal stage, there is a decreasing in estrogen levels, then it will stimulate unbalanced bone remodeling activity as the impact osteoblasts can not compensate for osteoclast work. By looking at previous studies, it has been known that nicotine can increase osteoblast activity in vitro which potentially used as an alternative therapy for osteoporosis. This study aims to determine the effect of nicotine on serum calcitonin in osteoporosis models. This study used osteoporosis mice by ovariectomy given nicotine at a dose of 0.25 mg/KgBB, 0.5 mg/ KgBB and 0.75 mg/KgBB for seven weeks after ovariectomy. The results showed that there were differences in calcitonin levels in the dose treatment (ANOVA, p = 0.000), but there was no relationship between dose and calcitonin levels (Spearman p = 0.083, r = -0.353). However, it shows that calcitonin levels decrease with increasing dosage, this insignificant result is due to the highly invasive ovariectomy procedure which cause mice prone to infection and continue to die after surgery. The conclusion of this study is that nicotine administration tends to increase serum calcitonin levels, which can be seen from the increase in the average calcitonin with increasing doses.
