Abstract
FGF-2 merupakan agen penting dalam proses penyembuhan luka karena berfungsi sebagai pendorong proliferasi fibroblas, menginduksi angiogenesis, dan meningkatkan sintesis kolagen. Gelatin ikan patin mengandung asam amino arginin dan glutamin yang berperan dalam proliferasi sel yang dapat meningkatkan jumlah ekspresi FGF-2 sehingga penyembuhan luka menjadi cepat. Tujuan penelitian ini adalah mengetahui pengaruh pemberian gelatin ikan patin (Pangasius djambal) terhadap ekspresi FGF-2 pada luka pasca pencabutan gigi tikus putih (Rattus norvegicus). Penelitian ini merupakan penelitian eksperimental laboratoris menggunakan Randomized Post Test Only Control Group Design. Sampel dipilih secara acak dan dibagi menjadi 6 kelompok (n=4), yaitu kelompok kontrol yang tidak diberi gelatin ikan patin dan kelompok perlakuan yang diberi gelatin ikan patin kemudian didekaputasi pada hari ke-3, ke-5, dan ke-7. Jumlah ekspresi FGF-2 dihitung dalam sediaan histologis dengan pewarnaan imunohistokimia dan diamati menggunakan mikroskop cahaya perbesaran 400x dalam 10 bidang lapang pandang. Hasil uji One Way ANOVA menunjukkan terdapat perbedaan yang bermakna antara kelompok kontrol dan perlakuan secara keseluruhan (nilai p<0,05). Hasil rata-rata jumlah ekspresi FGF-2 menunjukkan kelompok perlakuan mengalami peningkatan secara bertahap dan mencapai puncaknya pada hari ke-7. Kesimpulan penelitian ini adalah pemberian gelatin ikan patin (Pangasius djambal) berpengaruh terhadap peningkatan ekspresi FGF-2 pada luka pasca pencabutan gigi tikus putih (Rattus norvegicus).
English Abstract
FGF-2 is the key roles in wound healing process by promoting fibroblast proliferation, inducing angiogenesis and increasing the synthesis of collagenase. The patin fish (Pangasius djambal) gelatine containing the amino acid arginine and glutamine which has an important roles to initiate proliferation cells so that the number of FGF-2 expressions is increased so that wound healing will be fast. The purpose of this study was to determine the effect of patin fish gelatine to the number of FGF-2 expression in wound after white rat’s (Rattus norvegicus) tooth extraction. This study was an experimental laboratory study using Randomized Post Test Only Control Group Design. The samples were randomly divided into 6 groups (n=4) that is the control group who were not given patin fish gelatine and the treatment group who were given patin fish gelatine and then decaputed in 3rd, 5th and 7th day. The number of FGF-2 expressions was calculated in 10 random fields of view from histological preparations with immunohistochemistry (IHC) staining. Observations using the light microscope with magnification 400x. The result of One Way ANOVA showed there were significantly different between the control and treatment groups (p value < 0.05). The mean result showed that FGF-2 expression was gradually increased in treatment group and the highest increase was found in the 7th day treatment group. The conclusion of this study was the patin fish (Pangasius djambal) gelatine has effect to increase the number of FGF-2 expression in wound after white rat’s (Rattus norvegicus) tooth extraction.
