Abstract

Stres merupakan penyebab masalah tidur dengan patofisiologi yang melibatkan peningkatan histamin otak yang dihasilkan oleh sel mast otak. Quercetin diketahui dapat mencegah degranulasi sel mast perifer dan diketahui dapat melintasi sawar darah otak. Buah mengkudu (Morinda citrifolia) memiliki kandungan quercetin yang cukup tinggi yaitu sekitar 0,21-0,75g/100g daging buahnya. Penelitian ini bertujuan untuk mengetahui efek dari pemberian ekstrak etanol mengkudu intraperitoneal terhadap degranulasi sel mast otak tikus (Rattus norvegicus) yang diperlakukan stres auditorik subakut berupa white noise berdurasi 4 jam per hari selama 14 hari. Penelitian ini menggunakan tiga puluh tikus jantan dibagi menjadi 5 kelompok: kontrol negatif, kontrol positif, perlakuan 1 (quercetin murni), perlakuan 2 (antihistamin), dan perlakuan 3 (ekstrak etanol mengkudu). Sel mast diidentifikasi dengan menggunakan pewarnaan toluidine blue pada region talamus Dari hasil pengamatan dan penghitungan degranulasi sel mast, didapatkan persentase sel mast yang terdegranulasi berbeda signfikan antara kelompok kontrol positif dan negatif, dan tidak ada perbedaan signifikan antara perlakuan 1 (quercetin murni) dan perlakuan 3 (ekstrak etanol mengkudu), tetapi keduanya berbeda signifikan dengan kelompok kontrol positif (p<0.05). Jadi dapat disimpulkan bahwa injeksi intraperitoneal ekstrak etanol mengkudu dapat menghambat degranulasi sel mast otak.

English Abstract

Stress is known to be the cause of sleeping disorder by increasing the brain histamine which is produced by brain mast cell. Quercetin is known to be able to prevent the degranulation of peripheral mast cells, and quercetin is also known to be able to cross the blood-brain barrier. Noni fruit (Morinda citrifolia) contain pretty high dose of quercetin approximately 0,21-0,75g/100g of its fresh weight. This research was set out to know the effect of ethanol extract of Noni fruit intraperitoneal injection on the brain mast cell degranulation of rat (Rattus norvegicus) after 4-h daily exposure to white noise stress for 14 days. This research used 30 male rats divided into 5 groups which are negative control group, positive control group, quecetin intraperitoneal injection group, antihistamine intraperitoneal injection group, and ethanol extract of Noni fruit intraperitoneal injection group. Mast cell were identified by using toluidine blue staining in thalamus. This research shows that the percentage of mast cell degranulation is significantly different between the negative control group and positive control group, then there is no significantly different between quecetin intraperitoneal injection group and ethanol extract of Noni fruit intraperitoneal injection group, but both is significantly different compared to positive control group (P<0.05). Thus, it can be concluded that intraperitoneal injection of ethanol extract of Noni fruit can inhibit the degranulation of brain mast cells.
