Abstract

Kunyit banyak dipilih sebagai agen antiinflamasi dan antikanker secara tradisional. Terdapat banyak produk herbal mengandung kunyit untuk mengatasi antiinflamasi atau meredakan nyeri. Senyawa yang paling berperan memberikan efek farmakologis yaitu kurkumin dalam kunyit tersebut. Perlu dilakukan validasi metode analisis kurkumin menggunakan UHPLC-MS/MS karena memiliki hasil pemisahan senyawa dan deteksi senyawa yang sangat baik. Serta perlu dilakukan monitoring kualitas dengan penetapan kadar kurkumin dalam sediaan herbal tersebut. Penelitian ini bertujuan untuk mengetahui validitas medote analisis kurkumin yang digunakan dan mengetahui kadar kurkumin dalam beberapa sediaan herbal guna memenuhi monitoring kualitas sediaan tersebut. Sampel dipilih dengan cara metode purposive sampling dengan beberapa kriteria tertentu. Dilakukan validasi metode analisis kurkumin menggunakan UHPLC-MS/MS dengan fase gerak gradien A (0,1% asam format dalam aquabidest) dan B (0,1% asam format dalam acetonitrile) serta fase diam yaitu Kinetex EVO C18 100oA. Metode analisis dinyatakan valid dengan memenuhi parameter validasi yaitu selektivitas, linieritas (r=0,9997), akurasi (% perolehan kembali = 98,63-101,76%), presisi (%RSD=0,38-5,7%), LOD (0,11μg/ml) dan LOQ (0,34μg/ml). Berikutnya dilakukan penetapan kadar kurkumin dalam 3 sampel sediaan herbal mengandung kunyit. Kadar yang didapatkan sampel serbuk (0,03%), sampel kapsul (4,78%), dan sampel cair (0,03%).

English Abstract

Turmeric (Curcuma longa) widely used as the main ingredient of some herbal medicines to relieve pain and inflammation. Curcumin is the main secondary metabolites of Curcuma longa that has also shown some therapeutic activities such as anti-inflammatory and anti-cancer agent. An efficient, sensitive and precise Ultra High Performance Liquid Chromatography – Mass Spectrometry (UHPLC-MS/MS) was developed to determine curcumin content in several herbal medicine contains turmeric. The method was validated for linearity, selectivity, accuracy, precision, LOD and LOQ. Thus, the method can be used to determine curcumin content in herbal medicine to control herbal medicine quality. Objective of this research is to validate the method of curcumin analysis and determine curcumin content in herbal medicine. Validation was done through curcumin analysis using UHPLC-MS/MS with the gradient mobile phase A (0.1% format acid in aquabidest) and B (0.1% format acid in acetinitrile), while the static phase was Kinitex EVO C18 100o A. The method was finally recognized as valid by fulfilling several validation parameters such selectivity showed 3,2 minute as Retention Time (RT) also 285 m/z and 177 m/z as quantifier and qualifier product mass of curcumin , linearity (r=0.9997), accuracy (% recovery = 98,63-101.76%), precision (%RSD=0,38-5,70%), LOD (0,11 μg/ml) and LOQ (0,34 μg/ml). Curcumin level in three samples was determined from the herbal medicines containing turmeric. It showed that each herbal medicines sample contain curcumin 0.03%, 4,78% and liquid 0.03% for powder, capsule and liquid dosage form respectively. This research showed that the proposed method may be useful as an accurate, effective and sensitive for quantitative determination of curcumin content.
