Abstract

Faktor lingkungan dan genetik berperan dalam terjadinya LES. Gen pengatur komplemen C4 memicu sel T dan sel B memproduksi autoantibodi dan membentuk kompleks imun yang terkumpul di organ yang memicu inflamasi melalui proses aktivasi komplemen dan makrofag, sehingga terjadi kerusakan di banyak organ. Pencegahan terjadinya LES adalah dengan menggunakan antiinflamasi. Salah satu contoh dari anti-inflamasi ialah ekstrak daun cocor bebek. Penelitian eksperimental laboratorik ini bertujuan mengetahui efek ekstrak etanol daun cocor bebek pada mencit LES dengan induksi pristane. Metode yang digunakan adalah posttest only controlled group design dengan total mencit 25 ekor. Mencit dibagi menjadi 5 kelompok yakni kontrol negatif, kontrol positif, perlakuan dosis 1 (10,5mg/kgBB/hari), dosis 2 (21mg/kgBB/hari) dan dosis 3 (42mg/kgBB/hari) secara oral pada mencit Balb/c. Data dianalisis dengan uji normalitas, uji homogenitas, uji komparasi Kruskal-Wallis dan post-hoc MannWhitney serta uji korelasi Spearman. Didapatkan perbedaan signifikan kadar SGPT pada kelompok kontrol positif dengan kelompok dosis 3 (p=0,008), kelompok dosis 1 dengan kelompok dosis 3 (p=0,016) dan kelompok dosis 2 dengan kelompok dosis 3 (p=0,016). Dapat disimpulkan bahwa ekstrak etanol daun cocor bebek (Bryophyllum pinnatum) dapat memperbaiki kadar SGOT dan SGPT pada mencit BALB/c model LES dengan induksi pristane.

English Abstract

Genetic and environmental factors play a role in the occurrence of SLE. C4 complement trigger T cells and B cells to produce autoantibodies and immune complexes that are accumulated in the organs, it triggers an inflammatory process by macrophages and complement activation, so the damage in many organ occured. The prevention of the occurrence of the SLE are using anti-inflammatory agent. One example of anti-inflammatory is Bryophyllum pinnatum leaves extract.This experimental research aims to find out the effect of Bryophyllum pinnatum leaves with ethanol extract on SLE Balb/c mice pristane induced. The method used posttest only controlled group design with a total of 25 mice. Mice are divided into 5 groups : negative control, positive control, first dose treatment (10.5mg/kgBW/day), second dose (21mg/kgBW/day) and third dose (42mg/kgBW/day) PO in Balb/c mice. The data are analyzed with normality test, homogenity test, Kruskal-Wallis comparisons test, post-hoc Mann-Whitney test and Spearman correlation test. Obtained significant difference levels of SGPT in the positive control group with a third dose group (p = 0.008), first dose groups with third dose groups (p=0,016) and second doses groups with third doses groups (p=0,016). It can be concluded that the Bryophyllum pinnatum leaves with extract ethanol can improve the levels of SGOT and SGPT in Balb/C mice SLE models with pristane induced.
