Abstract
Toxoplasmosis merupakan penyakit zoonosis, yakni penyakit pada hewan yang dapat ditularkan ke manusia yang disebabkan oleh Toxoplasma gondii. Ditemukan adanya hubungan antara serologi positif Toxoplasma gondii dan obesitas.Profilin adalah komponen membran dari Toxoplasma gondii yang mampu merangsang ekspresi IL-12 dan sitokin proinflamasi melalui ikatan dengan TLR- 11. Ditemukan bahwa peningkatan kadar profilin pada individu obese berhubungan dengan peningkatan IL-6 dan IL-12 yang merupakan petanda awal terjadinya disfungsi adiposit pada individu obesitas, walaupun dengan korelasi yang lemah,Pada kondisi obesitas didapatkan peningkatan produksi ROS yang berasal dari peningkatan respirasi mitokondria dan dari hilangnya elektron yang dihasilkan dalam rantai transpor elektron. Ketika obesitas terus berlanjut untuk waktu yang lama, sumber antioksidan dapat menurun, seperti aktivitas enzim SOD dan GSH. Berdasarkan latar belakang tersebut, diperlukan penelitian untuk mengetahui efek paparan profilin Toxoplasma gondii terhadap kadar GSH pada tikus Rattus Norvegicus strain wistar. Penelitian ini adalah eksperimental laboratoris dengan menggunakan true experimental-post test control group design only. Hasil uji Kruskal-Wallis (P=0.393;α=≥0.05) menunjukkan tidak ada perbedaan pengaruh signifikan akan paparan profilin toxoplasma gondii terhadap kadar GSH pada tikus rattus norvegicus strain wistar. Hasil uji Korelasi (PI=0,531;PII=0,546;PIII=0,523;PIV=0,680;α=<0.05) terhadap ke 4 kelompok perlakuan berdasarkan dosis injeksi menunjukkan tidak ada hubungan yang signifikan antara dosis injeksi terhadap kadar GSH.Dari percobaan tersebut, dapat disimpulkan bahwa paparan profilin Toxoplasma gondii tidak memiliki pengaruh signifikan terhadap kadar GSH .
English Abstract
Toxoplasmosis is a zoonosis disease that caused by Toxoplasma gondii. The correlation between Toxoplasma gondii seropositivity and obesity was found. Profilin is a membrane component of Toxoplasma gondii that can stimulate IL-12 and pro-inflammation cytokine through bonds with TLR-11. Increase of profilin level within obese individual was correlate with increase of IL-6 and IL-12 as an early biomarker of adiposite disfunction, eventhough have weak correlation. The increase of ROS production within obese individual caused by increase of mithocondria respiration and electron from chain transport electron. The activity of antioxidant, like SOD and GSH will decrease if obesity is occur in long timeThis research was conducted to determine the effect of exposure profilin Toxoplasma gondii to GSH levels in Rattus Norvegicus Strain Wistar rats. This research is a laboratory experimental study with true experimental design -post test control group design only. The result of kruskal wallis test (P=0.393;=>0.05) shows that there is no significance difference on profilin exposure to GSH levels in Rattus Norvegicus Strain Wistar rats. The result of correlation test (PI=0,531;PII=0,546;PIII=0,523;PIV=0,680;α=<0.05) on 4 group based on injection dose shows no significance result. From the experiment, it can be concluded that the exposure of Toxoplasma gondii profilin don’t have significance influence the level of total GSH level.
