Abstract
Indonesia memiliki insiden kematian yang tinggi terhadap penyakit Demam Berdarah Dengue. Penyebab DBD yaitu virus Dengue yang dibawa oleh vektor Aedes aegypti. Pengendalian vektor efektif menggunakan larvasida, tetapi larvasida sintetik seperti abate dapat menimbulkan resistensi sehingga memerlukan larvasida alami/biolarvasida sebagai pengganti abate. Kulit manggis mengandung senyawa flavonoid, saponin, dan tanin yang mampu membunuh larva Aedes aegypti. Tujuan penelitian ini adalah untuk mengetahui efek ekstrak etanol kulit manggis (Garcinia mangostana L.) sebagai biolarvasida pada larva nyamuk Aedes aegypti instar III melalui kerusakan midgut. Sebanyak 500 larva digunakan sebagai sampel dan dibagi menjadi 5 kelompok dengan 4 kali pengulangan. Kelompok perlakuan tersebut yaitu kontrol negatif (air sumur), kontrol positif (abate 1%), ekstrak 0,5%, ekstrak 1%, dan ekstrak 2%. Kematian larva diamati dalam 24 jam. Kerusakan midgut larva diamati dengan mikroskop cahaya. Senyawa flavonoid, saponin, dan tanin terbukti terkandung dalam ekstrak berdasarkan uji fitokimia. Larvicidal activity ekstrak etanol kulit manggis pada konsentrasi 2% (tertinggi) sebesar 100% dan nilai LC100-nya yaitu 1,934%. Hasil pengamatan mikroskop cahaya menunjukkan adanya kerusakan midgut pada kelompok ekstrak dengan konsentrasi 0,5%, 1%, dan 2% yang ditandai dengan terbentuknya vakuola-vakuola pada sitoplasma. Pada penelitian ini dapat disimpulkan bahwa ekstrak etanol kulit manggis memiliki pengaruh sebagai biolarvasida pada larva nyamuk Aedes aegypti instar III melalui kerusakan midgut.
English Abstract
Indonesia has a high incidence of death against Dengue Hemorrhagic Fever. DHF is caused by Dengue virus that carried by Aedes aegypti as the vector. Controlling vectors by using larvicide is effective, but synthetic larvicides such as abate can cause a resistance, so that it requires natural larvicides/biolarvicides instead of abate. Mangosteen peel contains flavonoids, saponins, and tannins which are able to kill Aedes aegypti larvae. The purpose of this study was to determine the effect of mangosteen peel ethanol extract (Garcinia mangostana L.) as a biolarvicide on third instar Aedes aegypti larvae through midgut damage. 500 larvae were used as the samples and were divided into 5 groups with 4 repetitions. The groups were negative control (well water), positive control (abate 1%), extract 0.5%, extract 1%, and extract 2%. Larval mortality was observed in 24 hours. Midgut damage was observed by a light microscope. Mangosteen peel ethanol extract was proven to contain flavonoids, saponins, and tannins based on phytochemical test. Larvicidal activity of mangosteen peel ethanol extract with 2% of concentration (highest) was 100% and LC100 value was 1,934%. The observation of light microscope showed midgut damages in extract groups with concentration of 0,5%, 1%, and 2%, characterized by the formation of vacuoles in cytoplasm. This study concluded that the mangosteen peel ethanol extract has an effect as a biolarvicide on third instar Aedes aegypti larvae through midgut damage.
