Abstract

Kanker payudara merupakan kasus kanker tersering nomer satu pada perempuan diantara 10 kanker lainnya menurut data Badan Registrasi Kanker tahun 2012. Doxorubicin adalah agen kemoterapi yang sering digunakan dalam pengobatan kanker payudara tetapi mengalami resistensi pada sel MCF-7. Oleh karena itu penelitian ini bertujuan untuk mengetahui pengaruh kombinasi Doxorubicin dan ekstrak etanol daun benalu mangga (Dendrophthoe pentandara (L).) terhadap efek sitotoksik sel MCF-7. Uji sitotoksik dilakukan dengan MTT (3- (4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) dengan pemberian kombinasi Doxorubicin dan ekstrak etanol daun benalu mangga (Dendrophthoe pentandra (L.)) sebesar 12,5 µg/ml, 25 µg/ml dan 50 µg/ml. Sempel di inkubasi selama 24 jam, kemudian dilanjutkan dengan Elisa. Persentase kematian sel menunjukkan kontrol negatif sebesar 1%, kontrol positif sebesar 8%, perlakuan 1 sebesar 12%, perlakuan 2 sebesar 27%, perlakuan 3 sebesar 93% dan ekstrak etanol daun benalu mangga sebesar 37%. Data persentase kematian sel dihitung menggunakan IC50 untuk mengetahui ukuran efektivitas penghambatan sebesar 50% terhadap perlakuan. Data dari uji sitotoksik kombinasi Doxorubicin dan etanol daun benalu mangga (Dendrophthoe pentandra (L.)) diperiksa menggunakan metode uji Kruskal Willis dan didapatkan data memiliki nilai signifikan (p < 0,05). Kesempulan dari penelitian ini adalah kombinasi Doxorubicin dan ekstrak etanol daun benalu mangga (Dendrophthoe pentandra (L.)) pada 20 µg/mL memiliki efek sitotoksik tertinggi dari semua perlakuan terhadap sel MCF-7 dan didapatkan nilai IC50 sebesar 29,563 µg/m

English Abstract

Breast cancer is the number one most common cancer among women among 10 other cancers according to the Cancer Registration Instituion. Doxorubicin is a chemotherapy agent often used in the treatment of breast cancer but has resistance to MCF-7 cells. Therefore, this study aims to determine the effect of combination of Doxorubicin and ethanol extract of mango mistletoes (dendrophtoe pentandara (L.)) on the cytotoxic effects of MCF-7 cells. The cytotoxic assay was performed by MTT (3-(4,5-dimethylthiazol-2-yl)-2,5- diphenyltetrazolium bromide) using combination of Doxorubicin and ethanol extract of mango mistletoes (Dendrophthoe pentandra (L.)) of 12,5 μg / ml, 25 μg / ml and 50 μg / ml. Sample were incubated for 24 hours ,and then followed by Elisa. The percentage of cell death showed negative control was 1%, positive control 8%, group 1 was 12%, group 2 was 27%, group 3 was 93% and ethanol extract of mango mistletoes was 37%. Death cell percentage data was calculated using IC50 to find out the effective size of 50% inhibition on treatment. Data from cytotoxic test of combination of Doxorubicin and ethanol extract of mango mistletoes (Dendrophthoe pentandra (L.)) were examined using Kruskal Wallis test and obtained data that has significant (p <0,05). The conclusion of this research was the combination of Doxorubicin and ethanol extract of mango mistletoes (Dendrophthoe pentandra (L.)) at 20 μg/mL had the highest cytotoxic effect of all treatment on MCF-7 cells and obtained IC50 value of 29,563 µg/mL
