Abstract
Luka bakar derajat IIB mempunyai prevalensi 73% dan beresiko memicu inflamasi kronis akibat peningkatan jumlah makrofag berlebihan, sehingga perlu dimodulasi melalui perawatan luka. Salah satunya dengan daun cincau hijau. Penelitian ini bertujuan mengetahui pengaruh ekstrak daun cincau hijau (Cyclea barbata Miers) terhadap peningkatan jumlah makrofag blok waktu 4 jam, 48 jam, dan 72 jam pasca luka bakar derajat IIB pada tikus putih. Metode penelitian adalah true-experimental menggunakan RCBD dengan 36 ekor tikus putih galur wistar (n=4) yang dibagi dalam 3 blok waktu pembedahan (4 jam, 48 jam, dan 72 jam), kemudian tiap blok dibagi 3 subkelompok perawatan, yaitu kontrol negatif (NaCl 0,9%), kontrol positif (hidrogel), dan perlakuan (ekstrak daun cincau hijau 45%). Berdasarkan analisis, data terdistribusi normal dan sampel homogen (p > 0,05). Hasil Two-Way ANOVA menunjukkan perawatan dan waktu berbeda signifikan p= 0,000 terhadap peningkatan jumlah makrofag (p < 0,05). Dilanjutkan uji Post Hoc Tukey perawatan menunjukkan hidrogel dibandingkan ekstrak daun cincau hijau 45% tidak berbeda signifikan p= 0,146 dan NaCl 0,9% dibandingkan hidrogel dan ekstrak daun cincau hijau 45% berbeda signifikan p= 0,000 (p < 0,05). Sedangkan, uji Post Hoc Tukey blok waktu menunjukkan 48 jam dibandingkan 72 jam tidak berbeda signifikan p= 0,620 dan 4 jam dibandingkan 48 jam dan 72 jam berbeda signifikan p= 0,000 dan p= 0,002 (p < 0,05). Dapat disimpulkan bahwa ekstrak daun cincau hijau (Cyclea barbata Miers) 45% berpengaruh terhadap peningkatan jumlah makrofag pada blok waktu 4 jam, 48 jam, dan 72 jam pasca luka bakar derajat IIB pada tikus putih (Rattus norvegicus Strain Wistar).
English Abstract
Grade IIB burn has prevalence about 73% and has risk to stimulate the chronic inflammation cause increasing the number of macrophages excessively, so that the number of macrophages need to modulating with wound care. One of the wound care which can use is green cincau leaves. The aim of this study was to knew about the effects of green cincau leaves extract (Cyclea barbata Miers) toward increasing the number of macrophages in 4 hours, 48 hours, and 72 hours post grade IIB burn in white rats. The method was true-experimental with RCBD that used 36 white rats wistar strain (n=4) divided into 3 time blocks of surgery (4 hours, 48 hours, and 72 hours) and then divided into 3 subgroups of treatment, like negative control (NaCl 0,9%), positive control (hydrogel), and intervention (green cincau leaves 45%). Based on analysis, the data had distributed normally and the sample was homogen (p > 0,05). The result of Two-Way ANOVA showed that treatment and time blocks had significant differences to increasing the number of macrophages with p value= 0,000 (p < 0,05). Then, the test could be to continued with Post Hoc Test Tukey about the treatment subgroups which showed that there wasn’t significant differences between hydrogel toward green cincau leaves extract 45% with p value= 0,146 and there was significant differences between NaCl 0,9% toward hydrogel and green cincau leaves extract 45% with p value= 0,000 (p < 0,05). Meanwhile, Pos Hoc Test Tukey about time blocks of surgery showed that there wasn’t significant differences between 48 hours and 72 hours ith p value= 0, 620 and there was significant differences between 4 hours toward 48 hours and 72 hours with p value= 0,000 and p= 0,002 (p < 0,05). This study can be concluded that green cincau leaves 45% (Cyclea barbata Miers) has effects to increase the number of macrophages in time blocks, between 4 hours, 48 hours, and 72 hours post grade IIB burn in white rats (Rattus norvegicus Strain Wistar).
