Abstract

Lupus erythematosus systemik (LES) adalah penyakit multisistem yang disebabkan oleh produksi antibodi dan komposisi kompleks imun komposter komplemen yang mengakibatkan kerusakan jaringan. Meskipun patofisiologi yang tepat dari penyakit ini masih kurang dipahami, hal itu diketahui terkait dengan predisposisi genetik dan dipengaruhi oleh jenis kelamin dan lingkungan. Karena secara potensial banyak antibodi yang berbeda dapat diproduksi pada pasien SLE, sasaran spesifik organ yang berbeda dari antibodi ini dapat menyebabkan spektrum presentasi klinis yang luas, yang ditandai dengan remisi dan eksaserbasi. Dalam penelitian ini, jumlah sel T naif dengan penanda CD45, diamati saat menjalani imunoterapi spesifik dosis meningkat menggunakan self-antigen dsDNA pada mencit balb / c yang diinduksi pristane. Jumlah sel T naif menggunakan marker CD45 diukur dengan menggunakan flow cytometry menggunakan spleen mencit balb / c. Percobaan dilakukan di Laboratorium Biomedik, Laboratorium Farmakologi dan Laboratorium Parasitologi Fakultas Kedokteran Universitas Brawijaya. Dari percobaan tersebut, dapat disimpulkan bahwa jumlah sel T naif tidak menurun setelah peningkatan dosis self-antigen dsDNA seperti yang diperkirakan dalam hipotesis.

English Abstract

SLE is a disease that is multisystem which is caused by production of antibodies and complement-fixing immune complex deposition that eventually causes damage in tissue. Although the exact pathophysiology of the disease is still poorly understood it is known to be associated with genetic predisposition and influenced by gender as well as environment. Many different antibodies that are potentially produced in SLE patients can have different specific organ target whose antibodies can cause clinical presentations of wide spectrum that are characterized by exacerbations and remissions. In this research, the amount of naïve T cell with the marker CD45, is being observed while undergoing escalating dose specific immunotherapy using self-antigen dsDNA in pristane induced balb/c mice. The amount of naïve T cell using marker CD45 was measured using flow cytometry using balb/c mice spleen. The experiment was conducted at the Biomedical lab, Pharmacology Lab and Parasitology Lab of Medical Faculty University of Brawijaya. From the experiment, it can be concluded that the amount of naïve T cell did not decrease after escalating dose of self-antigen dsDNA as was predicted in the hypothesis
