Abstract

Kanker serviks merupakan salah satu jenis kanker penyebab kematian tertinggi bagi kaum wanita Indonesia. Hal ini menunjukan bahwa berbagai upaya pengobatan yang telah dilakukan seperti kemoterapi, terapi imunologik, dan operatif belum cukup efektif dalam menanggulanginya, sehingga perlu upaya lain dalam pengobatan kanker serviks tersebut. Senyawa 4-allil-2-metoksifenol atau disebut juga eugenol telah terbukti dapat menghambat pertumbuhan tumor dan memperlihatkan pengaruhnya yang signifikan dalam menghambat pertumbuhan sel kanker. Untuk meningkatkan kemampuan antiproliferatif eugenol dapat dilakukan modifikasi terhadap gugus allil eugenol menjadi gugus karbonil melalui reaksi adisi yang menghasilkan senyawa ester eugenol. Pada penelitian ini, dilakukan uji perbandingan antiproliferatif eugenol dan ester eugenol terhadap kultur sel HeLa kanker serviks yang diberikan delapan jenis perlakuan yang terdiri dari kelompok kontrol negatif yaitu kultur sel HeLa yang tidak diberi perlakuan, kelompok kontrol pelarut yaitu kultur sel HeLa yang diberikan DMSO, kelompok perlakuan 1, 2, dan 3 yaitu kultur sel HeLa yang masing-masing diberikan eugenol dengan konsentrasi 0.01%, 0.1%, dan 1%, dan kelompok perlakuan 4, 5, dan 6 yaitu kultur sel HeLa yang masing-masing diberikan ester eugenol dengan konsentrasi 0.01%, 0.1%, dan 1%. Kemudian, sel HeLa yang hidup maupun yang mati dianalisis menggunakan instrumen flow cytometry untuk menentukan tingkat apoptosis sel kultur. Hasil penelitian ini menunjukan ester eugenol memiliki tingkat antiproliferatif terhadap sel kanker serviks yang lebih tinggi dibandingkan dengan eugenol dengan nilai LC50 pada konsentrasi 48,73 ppm. English Abstract

Cervical cancer is one of the leading types of cancer cause of death in Indonesian women. This suggests that various treatment efforts such as chemotherapy, immunologic, and operative therapy have not been effective enough to overcome it, so it needs another innovation in the treatment of cervical cancer. The compound of 4-allyl-2-methoxyphenol or also called as eugenol has been shown to inhibit tumor growth and seen significant effect in inhibiting the growth of cancer cells. Improving the antiproliferative ability of eugenol can be done by modification of allyl eugenol groups into carbonyl groups by addition reactions which produce eugenol esters. In this study, the antiproliferative comparation of eugenol and eugenol ester were tested in cervical cancer cell heLa cultures which were given eight treatment types consisting of negative control group ie untreated HeLa cell culture, solvent control group ie HeLa cell culture given by DMSO, 1, 2, and 3 treatment of HeLa cell culture each of which were given eugenol with concentration of 0.01%, 0.1%, and 1%, and the treatment group 4, 5, and 6 were HeLa cell cultures, respectively given eugenol ester with concentration of 0,01%, 0,1%, and 1%. Then, the living or dead HeLa cells were analyzed using the flow cytometry instrument to determine the apoptotic cell culture level. The results of this study showed that eugenol ester had higher antiproliferative level against cervical cancer cell than eugenol with the value of LC50 at 48,73 ppm. This suggests, eugenol ester is quite effective in preventing the development of cervical cancer cells compared to eugenol.
