Abstract
Diabetes melitus tipe 2 (DMT2) adalah penyakit metabolik kronis yang di tandai dengan resistensi insulin. Resistensi insulin menyebabkan hiperglikemia mengakibatkan komplikasi seperti microangiopathy dan macroangiopathy. Sistem imun memproduksi IL-10 sebagai sitokin anti-inflamasi yang berperan sebagai imunostimulator dan imunosupresan pada sistem organ. Penelitian ini bertujuan menginvestigasi profil gen IL-10 dan ekspresi protein IL-10 pada organ ( ileum, otak, jantung, ginjal, hati, lambung dan paru-paru) tikus (Rattus norvegicus) model DMT2. Penelitian ini menggunakan 3 tikus jantan pada kelompok DMT2 dan 3 tikus jantan kelompok kontrol. Metode penelitian, isolasi, amplifikasi dan sekuensing DNA menggunakan primer IL-10. Profil pita protein menggunakan Experion pro-260 dan dot bloting. Hasil penelitian menunjukan adanya perbedaan profil gen IL-10 pada kelompok kontrol dan DMT2. Pada kelompok DMT2 ditemukan delapan mutasi pada organ otak dan dua puluh tujuh pada organ lambung. Profil pita protein dari kelompok kontrol dan DMT2 berbeda, tingkat ekspresi IL-10 kelompok DMT2 pada organ ileum, otak dan hati sangat tinggi. Kesimpulan dari penelitian ini, hewan model DMT2 menunjukan adanya mutasi pada organ otak dan lambung serta menginduksi meningkatnya tingkat ekspresi protein IL-10 pada ileum, otak dan hati
English Abstract
Type 2 diabetes mellitus (T2DM) is a metabolic disease cause by insulin restitance. Insulin restitance cause hyperglicemia resulting complication macroangiopathy and microangiopathy. IL-10 as a potential anti-inflamation to improve the function and restore organ function. This study aim to determine profil gen IL-10 and expression IL-10 protein in vary organs DMT2 rat model. This research was used six male rats Rattus norvegicus strain wistar divided into two gorups, control group (C) and T2DM group (DM) during two months hypercholesterol, injected STZ 25mg/KgBB after one week hypercholesterol. The DNA isolated from rat organ (ileum, brain, heart, kidney, liver, gastric and lung). The DNA was amplified and sequenced by primer IL-10 gene and then the IL-10 gene sequence was allignment by Bioedit v 7.2. The expression IL-10 protein of rat tissues was analyzed using Experion-Pro260 gel and dot blotting. In this study, we found some mutation in T2DM group of brain (eight mutations) and gastric (twenty-seven mutations) compared to other tissues of both groups. Intersetingly, these mutations may control the IL-10 protein expression which is the lowest of IL-10 in gastric against with IL- 10 brain (higher). In T2DM group, the level expression of IL-10 protein of ileum, brain and liver was hinger than other tissues.
