Abstract

Kelor (Moringa oleifera Lam.) merupakan tanaman berpotensi di bidang pertanian sebagai Zat Pengatur Tumbuh (ZPT) maupun sebagai antimikroba karena memiliki kandungan flavonoid, tanin, glikosida, dan terpinoids. Penelitian ini bertujuan untuk mengetahui daya hambat ekstrak daun kelor (Moringa oleifera) terhadap jamur A.solani yang di isolasi dari daun tomat yang terserang penyakit bercak daun dengan gejala yang khas bercak berwarna coklat dan berbentuk konsentris menuju pusat bercak. isolat yang didapatkan selanjutnya diidentifikasi morfologinya secara makroskopis maupun mikroskopis. Sampel positif A.solani dikultur dan dipergunakan pada perlakuan uji in vitro maupun in vivo. Pengujian terhadap persentase penghambatan ekstrak daun kelor (Moringa oliefera Lam.) dilakukan dengan menggunakan rancangan acak lengkap dengan 6 perlakuan yaitu kontrol positif (Mankozeb 80%), kontrol negatif (aquades) dan konsentrasi ekstrak yang berbeda (5%, 10%, 20% dan 30%,). Pengujian persentase penghambatan dilakukan dengan metode food poison (dicampur pada media tumbuh), kemudian dilakukan pengukuran berat kering miselium jamur. Perlakuan in vivo dilakukan dengan menginokulasi A. solani pada buah tomat muda dan diukur diameter gejala dan Lethal Concentration (LC50) nya. Besarnya daya hambat, berat kering dan diameter gejala penyakit dari masing-masing sampel dianalisis dengan analisis ragam sedangkan nilai LC50 dihitung menggunakan analisis regresi. Hasil penelitian menunjukkan bahwa daun kelor konsentrasi 5.000ppm mampu menghambat pertumbuhan jamur A. solani dengan nyata (F hitung > F tabel 5%), dengan hambatan paling optimum pada konsentrasi 30.000ppm dengan persentase penghambatan 100%, berat kering miselium seberat 0,0125 gram dan diameter gejala penyakitnya sebesar 0,2 cm. Untuk nilai LC50 ekstrak daun kelor didapatkan pada konsentrasi 10.555ppm

English Abstract

Moringa oleifera Lamk. is a potential crop in agriculture as a plant growth regulation and as an antimicrobial because it contains flavonoids, tannins, glycosides, and terpinoids. This study aims to determine the inhibitory power of Moringa oleifera leaves on A. solani patogen which were isolated from tomato leave attacked by early blight disease with typical symptoms of brown spots and concentric shaped towards the center of the spot. The isolates then identified by macroscopically and microscopically morphology. Positive samples of A.solani were cultured and used in both in vitro and in vivo treatments. Tests on inhibitory percentage of Moringa oliefera Lam. leaf extract were carried out using a completely randomized design with 6 treatments with positive control (Mankozeb 80%), negative control (aquades) and different extract concentrations (5.000ppm, 10.000ppm, 20.000ppm and 30.000ppm,). Inhibition percentage testing and Lethal Concentration 50 (LC50) is done by the food poisoning treatment (mixed on growing media), then measurements of dry weight of fungal mycelium are carried out. In vivo treatment was carried out by inoculating A. solani on young tomatos and measured the symptom of. The amount of inhibition, dry weight and diameter of the disease symptoms of each sample were analyzed by variance analysis while the LC50 values were calculated using probit regression analysis. The results showed that the concentration of 5.000ppm Moringa leaf was able to inhibit the growth of A. solani fungi significantly (F count> F table 5%), with the most optimum resistance at a concentration of 30.000ppm with a percentage of inhibition of 100%, dry weight of mycelium weighing 0.0125 grams and the diameter of the disease symptoms is 0.2 cm. For LC50 values, Moringa leaf extract was obtained at a concentration of 10.555ppm.
