Abstract

Acute Renal Failure (ARF) merupakan kelainan yang ditandai penurunan homeostasis ginjal dengan cepat disebabkan akumulasi toksin. Streptokinase digunakan untuk induksi ARF melalui peningkatan radikal bebas (ROS). Kulit manggis mengandung zat aktif xhanton yang menghambat produksi ROS dan menstimulasi regenerasi melalui growth factor. Penelitian bertujuan mengetahui pengaruh ekstrak kulit manggis terhadap ekspresi E-cadherin dan histopatologi ginjal tikus model ARF hasil induksi streptokinase. Penelitian menggunakan Rancangan Acak Lengkap menggunakan 20 tikus jantan berumur 12 minggu yang dibagi menjadi lima kelompok: kontrol negatif, kontrol positif (tikus diinduksi streptokinase 6000IU) dan kelompok terapi dengan dosis ekstrak kulit manggis: 200 mg/kgBB (T1); 400 mg/kgBB (T2); dan 600 mg/kgBB (T3). Ekspresi Ecadherin diamati menggunakan metode imunohistokimia dan dihitung dengan Immunomembrane. Data dianalisa dengan uji ragam One-way ANOVA dan uji BNJ menggunakan SPSS for windows 17.0. Hasil penelitian menunjukkan bahwa pemberian terapi ekstrak kulit mangis pada hewan model ARF secara signifikan meningkatkan E-cadherin pada T1, T2 dan T3. Hal ini didukung pengamatan histopatologi yang menunjukkan penurunan akumulasi sel polimorfonuklear, dan adanya regenerasi korpuskula ginjal pada T1, T2 dan T3 dibandingkan kelompok kontrol positif. Berdasarkan ekspresi E-cadherin dan histopatologi ginjal, dosis ekstrak kulit manggis 400 mg/kgBB menunjukkan dosis terapi optimum pada tikus model ARF

English Abstract

Acute Renal Failure (ARF) is a syndrome characterized by rapid loss of kidney excretory function caused by toxins accumulation. Streptokinase used to induce ARF by increasing of free radical output (ROS). Mangosteen pericarp consists of xanthones which can inhibit ROS production and stimulate tissue regeneration by growth factor. This research was conducted to assess the effects of mangosteen pericarp extract (Garcinia mangostana) on renal E-cadherin expression and histopathology of streptokinase-induced ARF rats. This research used completely randomized design. Twenty adult male rats at three months of age were divided into five groups: negative control; positive control (rats were intravenously injected by streptokinase 6000IU); and treatment groups with different doses of mangosteen extract were 200 mg/kgBW (T1); 400 mg/kgBW (T2); and 600 mg/kgBW (T3). E-cadherin expression were observed by immunohistochemistry and quantified by Immunomembrane program. Data were analyzed by One-Way Analysis of Variance (ANOVA) (α<0.05) by using SPSS 17.0 for Windows. Mangosteen pericarp extract significantly increased E-cadherin expression in T1, T2 and T3. Histopathology examination showed tissues regeneration and decreased of polimorphonuclear cell accumulation in T1, T2 and T3 between positive groups.
