Abstract
Oviductin merupakan salah satu jenis protein yang disekresikan pada saluran reproduksi kambing peranakan Ettawah dengan berat molekul spesifik antara 55 – 65 kDa. Protein ini, secara umum, memiliki peranan untuk membantu keberhasilan fertilisasi dengan mencegah terjadinya polispermia melalui mekanisme vitelline block serta membantu proses perkembangan awal embrio. Penelitian ini bertujuan untuk mengetahui ekspresi oviductin pada ovarium kambing fase luteal dan folikuler serta mengetahui perbedaan jumlah folikel di ovarium. Sampel ovarium yang digunakan berasal dari limbah Rumah Potong Hewan. Analisa ekspresi oviductin dilakukan menggunakan metode imunohistokimia dan perhitungan jumlah folikel ovarium dilakukan menggunakan preparat dengan pewarnaan Hematoxylin Eosin (HE). Ekspresi oviductin pada preparat dengan metode imunohistokimia kemudian dianalisa secara kuantitatif menggunakan immunoratio dan dilanjutkan dengan uji t independent. Hasil penelitian menunjukkan bahwa nilai rata- rata ekspresi oviductin pada fase folikuler ovarium adalah sebesar 89% dan pada fase luteal adalah sebesar 12%. Sementara itu rata – rata jumlah folikel ovarium pada fase folikuler lebih banyak dibandingkan fase luteal. Disimpulkan bahwa jumlah folikel ovarium berhubungan dengan peranan oviductin dalam folikulogenesis
English Abstract
Oviductin is kind of protein secreted in the goat reproductive tract, include peranakan ettawah goat, with specific molecular weight of 55 – 65 kDa. This protein is, in general, have a role to help the success of fertilization by preventing the occurrence of polispermia through the mechanism of the vitelline block and help the process of the development of the early embryo. Research purpose was determine the expression of oviductin in the luteal and follicular phase of goat and differences the number of follicles in the ovaries. Ovarian samples were obtained from slaughterhouse wastes. Oviductin expression were analyzed by immunohistochemistry technique and the number of ovarian follicle evaluated by Hematoxylen Eosin (HE staining). The results showed that oviductin expression in ovarian follicular phase and luteal phase are 89% and 12%, respectively. Meanwhile, the average number of ovarian follicles in the follicular phase is more than the luteal phase. It was concluded that the number of ovarian follicles is related to the role of oviductin in folliculogenesis.
