Abstract

Buaya Senyulong (Tomistoma schlegelii) merupakan jenis buaya air tawar yang dilindungi keberadaannya di Indonesia. Penelitian mengenai analisa kekerabatan antar individu buaya Senyulong menggunakan DNA mitokondria (mtDNA) pada gen Cytochrome C Oxidase sibunit 1 (COI) perlu dilakukan demi mendukung upaya pelestariannya. Gen COI berukuran 479 bp diperoleh dari 3 sampel darah buaya Senyulong setelah melalui tahap isolasi DNA dan amplifikasi dengan metode Polymerase Chain Reaction (PCR) menggunakan sepasang primer forward (HMD_F) 5’-GCT TTT CTC CGC TTT CAT CG -3’ dan reverse (HMD_R) 5’-CCC ATA TAC CCG AAT GGT TC-3’. Sekuens yang diperoleh dari amplifikasi gen COI dianalisa untuk memperlihatkan kekerabatan antar sampel individu (intraspesies), lalu dibandingkan dengan Gharial (Gavialis gangeticus) dan buaya Irian (Crocodylus novaeguineae) (interspesies) menggunakan program BioEdit®, Basic Local Aligment Search Tool (BLAST) dari NCBI, dan program Molecular Evolutionary Genetics Analysis (MEGA). Hasil penelitian menunjukkan bahwa kekerabatan buaya Senyulong yang berada di Predator Fun Park (PFP) kota Batu Jawa Timur berhasil dideskripsikan berdasarkan Gen COI (Cytochrome C oxidase subunit 1) secara intraspesies maupun interspesies. Jarak genetik intraspesies sampel buaya Senyulong yang dibandingkan dengan sekuen gen COI buaya Senyulong database NCBI bernilai sama atau kurang dari 2%. Jarak genetik interspesies yang dibandingkan dengan Gharial (familia Gavialidae) bernilai 13% sampai 14,3%, sedangkan jika dibandingkan dengan buaya Irian (familia Crocodylidae) bernilai 15,7% sampai 16,4%

English Abstract

Senyulong crocodyle (Tomistoma schlegelii) is a freshwater crocodile protected by its existence. Research on the analysis of relationship between individual Senyulong crocodyle using mitochondrial DNA (mtDNA) in the Cytochrome C Oxidase sibunit 1 (COI) gene needs to be done in order to support conservation efforts. The 479 bp COI gene was obtained from 3 samples of Senyulong crocodyle blood after passing through the DNA isolation and amplification stages using Polymerase Chain Reaction (PCR) with forward primer (HMD_F) 5'-GCT TTT CTC CGT TTT CG CAT -3 ' and reverse ( HMD_R) 5'- CCC ATA TAC CCG AAT GGT TC-3 '. Sequences obtained from COI gene amplification are analyzed for attempted kinship rates between individual samples (intraspecies), then compare with the Gharial (Gavialis gangeticus) and the New Guinea crocodyle (Crocodylus novaeguineae) (interspesies) using BioEdit®, Basic Search Aligment Search Tool (BLAST) from the NCBI, and the Molecular Evolutionary Genetics Analysis (MEGA) program. The results showed that Senyulong crocodyle relationship level located in Predator Fun Park (PFP) Batu city of East Java was successfully described by Genes COI (Cytochrome C oxidase subunit 1). The genetic distance intraspecies of Senyulong crocodyle samples compared with gene sequences of Senyulong crocodyle NCBI databases is equal or less than 2%. The genetic distance of the interspesies compared with the Gharial (Gavialidae family) is 13% to 14.3%, whereas when compared to Irian crocodyle (Crocodylidae family) is 15.7% to 16.4%.
