Abstract
Gagal ginjal akut merupakan gangguan dimana ginjal tidak dapat melakukan fungsi normalnya dalam pembentukan urin dalam waktu yang cepat. Salah satu penyebab gagal ginjal akut adalah antibiotik gentamisin. Gentamisin dapat menyebabkan kerusakan sel pada tubulus nefron. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian gentamisin dosis bertingkat berdasarkan gambaran histopatologi ginjal dan ekpresi Tumor Necrosis Factor Alpha (TNF-α) ginjal pada hewan model gagal ginjal akut. Hewan coba dalam penelitian ini adalah 20 ekor tikus ( Rattus norvegicus ) jantan dengan berat 200g berumur 2 bulan yang dibagi menjadi 4 kelompok. Kelompok 1 adalah kelompok kontrol negatif yang tidak diberi perlakuan. Kelompok 2,3 dan 4 adalah kelompok yang diinduksi gentamisin dengan dosis 30mg/kgBB, 40mg/kgBB, dan 50mg/kgBB. Kerusakan ginjal yang disebabkan oleh gentamisin diamati melalui preparat histopatologi ginjal menggunakan pewarnaan Hematoxylen Eosin (HE) dan kadar ekspresi TNF-α dengan flowcytofotometry. Analisis data secara deksriptif untuk hitopatologi ginjal sedangkan untuk kadar ekspresi TNF-α menggunakan uji one way ANOVA dan uji lanjutan BNJ dengan tingkat kepercayaan 95% (α=5). Hasil dari penelitian ini adalah terjadi kerusakan pada ginjal pada semua kelompok perlakuan dosis gentamisin berupa nekrosis di tubulus kontortus, kapsula bowman dibandingkan dengan kelompok kontrol negatif. Kadar TNF-α dosis 40mg/kg BB mengalami peningkatan dibandingkan dengan kelompok kontrol negatif. Kesimpulan dari penelitian ini adalah kelompok perlakuan dengan dosis 40mg/kg BB dapat menyebabkan kerusakan ginjal dan peningkatan kadar TNF- α dibandingkan dengan kontrol negatif
English Abstract
Acute kidney failure is a disorder in which the kidneys cannot perform their normal function in the formation of urine in a fast time. One of the causes of acute renal failure is antibiotics gentamicin. Gentamicin can cause cell damage to the tubule nephron. This study aims to determine the effect of multilevel gentamicin administration based on renal histopathology and renal expression of Necrosis Factor Alpha (TNF-α) in animal models of acute renal failure. The experimental animals in this study were 20 male rats (Rattus norvegicus) weighing 200g 2 months old which were divided into 4 groups. Group 1 is a negative control group that is not treated. Groups 2,3 and 4 were the group induced by gentamicin at a dose of 30 mg/kg BW, 40 mg/kg BW, and 50 mg/kg BW. Kidney damage caused by gentamicin was observed through renal histopathology preparations using Hematoxylen Eosin (HE) staining and TNF-α expression levels with flowcytofotometry. Descriptive data analysis for renal histopatology while TNF-α expression levels used one way ANOVA test and BNJ follow-up test with 95% confidence level (α = 5). The results of this study were kidney damage in all gentamicin dose treatment groups in the form of necrosis in the contour tubules, capsule bowman compared to the negative control group. TNF-α levels of 40 mg/kg BW have increased compared to the negative control group. The conclusion of this study is that the treatment group with a dose of 40 mg/kg BW can cause kidney damage and increase levels of TNF-α compared with negative controls
