Abstract
Hiperkolesterolemia merupakan suatu keadaan dimana terjadi peningkatan kadar kolesterol di dalam plasma darah melebihi kadar normal yang ditandai dengan meningkatnya kadar LDL, trigliserida, dan kolesterol total. Angkak dapat digunakan sebagai obat anti kolesterol karena mengandung lovastatin yang mampu menghambat HMG-CoA reduktase, yaitu enzim yang mengontrol jalur biosintesis kolesterol di dalam hati. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian angkak terhadap kadar malondialdehida (MDA) dan gambaran histopatologi jantung pada tikus model hiperkolesterolemia. Hewan coba yang digunakan adalah tikus putih (Rattus norvegicus) jantan usia 10-12 bulan dengan berat 150-200 gram. Tikus dibagi menjadi 5 kelompok, yaitu kelompok kontrol negatif, kelompok kontrol positif, kelompok P1, P2, P3 terapi angkak dengan dosis 0,5 g/ekor/hr, 1 g/ekor/hr, dan 1,5 g/ekor/hr. Pemberian terapi dilakukan pada hari ke 15 setelah induksi pakan Hiperkolesterol. Kadar MDA diukur menggunakan metode TBA (Thiobarbituric Acid) dan gambaran histopatologi jantung dengan pewarnaan HE. Hasil penelitian menunjukkan bahwa terapi angkak secara signifikan (p<0,05) menurunan kadar MDA. Pada terapi dosis 1 g/ekor/hari adalah dosis efektif dalam menurunkan kadar MDA sebesar 33,4 %. Gambaran histopatologi jantung menunjukkan adanya perbaikan kardiomiosit yang ditunjukkan dengan perubahan struktur kardiomiosit yang bergelombang dan tidak beraturan menjadi lebih baik seperti keadaan normal. Dapat disimpulkan bahwa terapi angkak mampu menurunkan kadar MDA dan memperbaiki gambaran histopatologi jantung pada tikus model hiperkolesterolemia.
English Abstract
Hypercholesterolemia is a condition when cholesterol levels in blood plasma increased and exceeded the normal range marked by the increase of LDL, triglyceride and total cholesterol. Red yeast rice (angkak) could be used as an anticholesterol because it contains lovastatin that could inhibit the reductase of HMGCoA, which is an enzyme that controls biosynthesis path of cholesterol in the liver. The purpose of this study was to find out the therapy effect of red yeast rice fermented by Monasus sp. on malondialdehyde (MDA) level and histopathological appearance of heart on rats (Rattus norvegicus) with hypercholesterolemia. The animals that were used consisted of male rats (Rattus norvegicus) age 10-12 months and weighed around 150 – 200 grams. The rats were divided into five groups, which are negative control group, positive control group, P1, P2 and P3 group that was given red yeast rice therapy with doses of 0.5 g/rat/day, 1 g/rat/day and 1,5 g/rat/day. The therapy was given on the 15th day after the induction of high cholesterol food diet. The MDA level was measured by TBA (Thiobarbituric Acid) method and histopathological appearance was stained by Hematoxylin-Eosin (HE). The result showed that red yeast rice significanty (p<0,05) decrease MDA levels unit effective therapy dose of 1 g/rat/day up to 33,4% level of MDA. Histopathological image of heart showed a recovery of cardiomyocyte that marked by the changes of wavy and irregular cardiomyocyte structure to normal cardiomyocytes structure. The conclusion of this study was red yeast rice could decrease MDA levels and recover histopathological appearance of heart on hypercholesterolemia rats.
