Abstract
Karies gigi merupakan penyakit infeksi yang menyebabkan kerusakan jaringan gigi hingga terbentuk kavitasi dan sering terjadi pada anjing. Penyebab utama karies gigi adalah Streptococcus mutans yang menyebabkan fermentasi karbohidrat, mengubah pH mulut menjadi asam, demineralisasi gigi, membentuk biofilm, dan bersifat imunosupresan. Kondisi pH mulut juga akan menurun drastis apabila diberi pakan diet kariogenik seperti sukrosa. Kefir susu kuda Sumbawa memiliki efek imunostimulator, kandungan antibakteri dan mineral untuk remineralisasi gigi. Tujuan penelitian ini adalah untuk menguji kemampuan kefir susu kuda Sumbawa dalam mencegah karies gigi terhadap produksi TNF-α – Gr1 dan IL-10 – TCD4 pada hewan coba yang diinduksi S. mutans. Penelitian ini eksperimental dengan metode post-test only control design menggunakan model Rancangan Acak Lengkap (RAL). Hewan coba yang digunakan adalah mencit Balb/c jantan, berat badan ± 20-30 gram, dan umur 1,5 bulan dengan jumlah sampel 20 ekor yang diberikan Ampicilin selama 3 hari untuk menekan pertumbuhan bakteri oral sebelum perlakuan, kemudian dibagi menjadi: kelompok K(-) sebagai kelompok kontrol negatif, kelompok K(+) sebagai kelompok model karies gigi yang diinduksi bakteri S. mutans 4 x 109 CFU/ekor setiap hari selama 5 hari dan diet sukrosa 5%, kelompok P1, P2, dan P3 sebagai kelompok preventif dengan masing-masing pemberian kefir susu kuda Sumbawa dosis 0,1 ml/ekor, 0,25 ml/ekor, dan 0,5 ml/ekor setiap hari selama 14 hari yang diberikan sebelum diinduksi S. mutans dan diet sukrosa 5%. Parameter yang diukur adalah produksi TNF-α – Gr1 dan IL-10 – TCD4 dari glandula saliva menggunakan metode flowcytometry. Data yang diperoleh dianalisis secara statistik menggunakan metode One-Way ANOVA dengan α=0,05. Hasil penelitian menunjukkan peningkatan produksi TNF-α – Gr1 dan penurunan produksi IL-10 – TCD4 secara optimal pada dosis 0,5 ml/ekor dengan signifikansi p<0,05. Kesimpulan dari penelitian ini adalah kefir susu kuda Sumbawa memiliki efek preventif untuk karies gigi.
English Abstract
Dental caries is an infectious disease that causes damage on tooth tissue to form cavitation and often occurs in dogs. The main cause of dental caries is Streptococcus mutans which causes carbohydrate fermentation, turning the pH of the mouth into acid, dental demineralization, forming biofilms, and immunosuppressant. Oral pH conditions will also decrease drastically when fed by a cariogenic diet such as sucrose. Sumbawa’s Horse Milk Kefir has immunostimulator effects, antibacterial and mineral contents for tooth remineralization. The objective of this study was to test the ability of Sumbawa’s Horse Milk Kefir in preventing dental caries against TNF-α – Gr1 and IL-10 – TCD4 production in S. mutans-induced susceptible animals. This research was experimental using completely Randomized Complete Design (RAL) model. The experimental animals use male Balb/c mice, weight ± 20-30 grams, and 1.5 months of age with 20 samples given by Ampicilin for 3 days to suppress the growth of oral bacteria prior to treatment, then divided into: K group (-) as a negative control group, group K (+) as a group of induced dental caries model of S. mutans 4 x 109 CFU/mice every day for 5 days and 5% sucrose diet, P1, P2 and P3 groups as group preventive treatment with each giving Sumbawa’s Horse Milk Kefir dose 0.1 ml / mice, 0.25 ml / mice, and 0.5 ml / mice every day for 14 days given before induced S. mutans and 5% sucrose diet. The parameters measured are the production of TNF-α – Gr1 and IL-10 – TCD4 from salivary glands using flowcytometry method. The data obtained analyzed statistically using One-Way ANOVA method with α=0,05. The results showed elevated levels of TNF-α – Gr1 and decreased IL-10 – TCD4 levels with optimal effect at dose of 0.5 ml/mice with a significance p<0.05. The conclusion of this study is that Sumbawa’s Horse Milk Kefir has a preventive effect on dental caries.
