Abstract
Penelitian bertujuan untuk meneliti profil glikoprotein, nutrisi, dan mikroba dari tiga jenis sarang burung walet asal Indonesia yakni sarang burung walet putih, sarang burung walet hitam, dan sarang burung seriti. Profil nutrisi dan mikroba dianalisis menggunakan rancangan acak lengkap dimana perlakuan (jenis sarang burung walet), sedangkan profil glikoprotein dianalisis secara deskriptif. Profil nutrisi dianlisis dengan metode proksimat dan kandungan glikoprotein dianalisis dengan Glycoprotein Estimation Kit. Profil mikroba dianalisis dengan metode angka lempeng total. Analisis E. Coli dan Salmonella menggunakan metode media selektif. Profil glikoprotein dilakukan menggunakan elektroforesis pewarna coomassie brilliant blue, elektroforesis pewarna glikoprotein dan LC-MS/MS. Hasil penelitian menunjukkan bahwa kandungan nutrisi tiga jenis sarang burung walet asal Indonesia berbeda pada setiap jenisnya. Hasil analisis Profil mikroba menunjukkan bahwa kandungan angka lempeng total tertinggi dimiliki oleh sarang burung walet hitam sedangkan yang terendah adalah sarang burung walet putih dan sarang burung walet seriti berada diantaranya. Kandungan E.coli dan Salmonella ketiga jenis sarang burung walet asal Indonesia adalah negatif. Hasil analisis profil glikoprotein dengan elektroforesis pewarna coomassie brilliant blue menunjukkan pita protein pada berat molekul yang berbeda yaitu 140,8; 64,8; 21,2 kDa untuk ekstrak air sarang burung walet putih, 27,1; 21,2; 15,2 kDa untuk ekstrak air sarang burung walet hitam dan 140,8; 116, 36,2 KDa untuk ekstrak air sarang burung walet seriti. Profil glikoprotein dengan elektroforesis pewarna glikoprotein menunjukkan hasil yang berbeda 140,8 dan 64,8 kDa untuk ekstrak air sarang burung walet putih, 140,8 dan 116,0 kDa untuk ekstrak air sarang burung walet seriti dan 28,4 dan 105,3 kDa untuk ekstrak air sarang burung walet hitam. Hasil analisis menggunakan LC-MS/MS menunjukkan kehadiran glikoprotein pada ekstrak air ii sarang burung walet putih namun pada ekstrak air sarang burung walet seriti dan sarang burung walet hitam tidak ditemukan. Hasil LC-MS/MS ini didukung oleh hasil elektroforesis pewarna coomassie brilliant blue dan pewarna glikoprotein yang menunjukkan kemungkinan glikoprotein berada pada ekstrak air sarang burung walet putih dan sarang burung walet seriti karena berat molekul karbohidrat (gula) hasil analisa pewarna gliikoprotein masih dalam kisaran berat molekul protein yang berikatan dengan karbohidrat (gula) hasil analisa pewarna coomassie brilliant blue. Hasil analisis profil glikoprotein juga menunjukkan bahwa profil glikoprotein sarang burung walet putih diduga berbeda dengan profil glikoprotein sarang burung walet hitam dan sarang burung walet seriti, namun perbedaan profil glikoprotein sarang burung walet hitam dan seriti belum diketahui.
English Abstract
The research aimed to study glycoprotein, nutrition and microorganisms from three kinds of Indonesian edible bird nest consisting of white edible bird nest, black edible bird nest and swallow edible bird nest. Nutrition and microorganism profiles were analyzed by using randomized complete design with treatments on the kinds of edible bird nest while glycoprotein profile was analyzed descriptively. Nutrition profile was analyzed using proximate method and glycoprotein was analyzed with Glycoprotein Estimation Kit. Microorganism profile was analyzed using total plate count method. E. Coli and Salmonella analysis was conducted by using selective medium. Glycoprotein profile was conducted by using coomassie blue staining electrophoresis, glycoprotein staining electrophoresis and LC-MS/MS. The results showed that nutrition from the three kinds of edible bird nest in Indonesia were different in each kind. Microorganisms profile analysis results showed that the highest total plate number was found in black edible bird nest while the lowest was found in white edible bird nest and the swallow edible bird nest was between them. E. Coli and Salmonella content in three kinds of edible bird nest from Indonesia were negative. The results on glycoprotein profile using coomassie brilliant blue staining electrophoresis showed that protein prepared by water extraction, band on various high intensity molecular weight were 140.8; 64.8; 21.2 kDa for the white edible bird nest`s, 27.1; 21.2; 15.2 kDa for black edible bird nest`s, and 140.8; 116; 36.2 kDa for swallow edible bird nest`s. Glycoprotein profile using glycoprotein staining showed different results, namely 140.8 and 64.8 kDa for water extract of white edible bird nest`s; 140.8 dan 116.0 kDa for swallow edible bird nest`s; 28.4 and 105.3 kDa for black edible bird nest`s. Analysis results using LC-MS/MS showed the presence of glycoprotein in the white edible bird nest`s water extract, but none was found in the water extract of black and the swallow edible bird nest`s. LC-MS/MS results were supported by the results of coomassie blue staining and the glycoprotein electrophoresis which showed the possibility of glycoprotein content in the white and the swallow edible bird nest`s prepared by water extraction since carbohydrate (sugar) molecular weight from glycoprotein iv staining analysis was still in the range of carbohydrate (sugar) bonded protein molecular weight resulted from coomassie brilliant blue staining analysis. Analysis result of glycoprotein profile showed that the glycoprotein profile of white edible bird`s nest was suspected different from black edible bird`s nest and swallow edible bird`s nest, but the differences of black and swallow edible bird nest were still unknown.
