Abstract
Latar Belakang. Transflutrin merupakan insektisida golongan pyrethroid yang bersifat lipofilik dan selama ini dianggap aman. Transflutrin dapat berinteraksi dengan reseptor estrogen. Sejumlah EDC ( endocrine disrupting chemicals ) dapat mengakibatkan obesitas dengan membuat kekacauan target sensor metabolik, di antaranya melalui reseptor estrogen. Kondisi obesitas menyebabkan meningkatnya kadar serum leptin dan menurunnya kadar serum adiponektin. Tujuan. Mengetahui pengaruh paparan sub-kronis senyawa insektisida transflutrin perinhalasi terhadap terjadinya obesitas pada tikus Rattus novergicus melalui peningkatan kadar leptin dan penurunan kadar adiponektin. Metode. Penelitian eksperimental in vivo pada 35 tikus jantan, terbagi dalam 5 kelompok. Kelompok I sebagai kontrol negatif, tanpa perlakuan. Kelompok II sebagai kontrol positif, mendapat paparan n-hexana sebagai pelarut. Kelompok III, IV dan V secara berturut-turut mendapat paparan n-hexana plus transflutrin 0,5 mg, 1 mg dan 2 mg. Kadar serum leptin dan adiponektin dinilai menggunakan metode quantikine immunoassay ELISA ( enzyme-linked immunosorbent assay ) kit dengan nomer katalog E0561Ra dan E0758Ra . Hasil. Kadar serum leptin meningkat pada kelompok yang mendapat paparan n-hexana dan paparan n-hexana plus transflutrin, namun kenaikan tertinggi didapatkan pada kelompok kontrol positif. Di antara kelompok yang mendapat paparan transflutrin, kenaikan kadar leptin sebanding dengan bertambahnya dosis transflutrin. Kadar serum adiponektin menurun pada kelompok yang mendapat paparan n-hexana plus transflutrin, yang berbanding terbalik dengan bertambahnya dosis transflutrin. Tidak didapatkan perbedaan pertambahan berat badan antar kelompok. Terdapat korelasi negatif yang signifikan antara kadar serum leptin dengan adiponektin (p=0,01). Kesimpulan. Pemberian paparan subkronis senyawa transflutrin per inhalasi meningkatkan kadar serum leptin dan menurunkan kadar serum adiponektin. Kadar serum leptin berhubungan terbalik dengan kadar adiponektin. Tidak didapatkan perbedaan pertambahan berat badan.
English Abstract
Background . Transflutrin, a lipophilic pyrethroid class of insecticides, is considered as a safe substance . Transflutrin can interact with estrogen receptor . Some endocrine disrupting chemicals ( EDCs) can caused obesity by disrupt target metabolic sensors , including through interaction with estrogen receptor . O besity causes increased levels of leptin and decreased levels of adiponectin . Objective . To d etermine sub - chronic obesogenic effect of transflutrin exposure in Rattus novergicus through inhalation by increased levels of leptin and decreased levels of adiponectin . Methods . In vivo experimental study on 35 male rats , divided into 5 groups . Group I as negative control ( no treatment) . Group II as positive control, exposed to n - Hexana as solvent. Group III, IV and V exposed to n - Hexana plus transflutrin 0.5 mg, 1 mg and 2 mg consecutively . Serum leptin and adiponectin was examined using quantikine immunoassay method ELISA kit (catalog no. E0561Ra and E0758Ra ) . Results . Leptin serum level increased in both solvent and solvent plus transflutrin group. However highest increased was detected in rat exposed to solvent. Among trasflutrin exposed rat, increased of leptin level were dose dependent. Serum adiponectin levels showed a downward trend which is inversely proportional with increase transflutrin doses . re is no weight gain difference among groups. T here was negative correlation between serum leptin and adiponectin levels ( p = 0.01 ). Conclusion .
