Abstract

Xylanases are extensively used in animal feed to breakdown arabinoxylans in ingredients of feed and degrading viscosity of raw substance. aim of this study was to investigate effect of dietary supplementation by B. amyloliquefaciens (amy wild type) and xylanaseexpressed recombinant B. amyloliquefaciens (amy R8) probiotics on health status and hepatic metabolism genes expression of zebrafish (D. rerio) and tilapia (O. niloticus), and tilapia growth performance. zebrafish and tilapia fishes were fed for 2 months with three different diets: control (without probiotic), amy wild type supplemented with 109 cfu mg-1 B. amyloliquefaciens, and amy R8 supplemented with 109 cfu mg-1 xylanaseexpressed recombinant B. amyloliquefaciens. In vitro and vivo assay showed iv that amy R8 significantly increased xylanase activity compared to amy wild type and control diets. Weight gain, feed efficiency, and specific growth rate (SGR) significantly increased in tilapia after feeding with amy wild type and amy R8 probiotic for 2 months. survival rates were increased in Aeromonas hydrophila and Streptococcus agalactiae infected zebrafish or A. hydrophila infected tilapia after feeding with amy wild type and amy R8 for 2 months. expression level of immune-related genes such as interleukin (IL)- 1β, IL-6, IL-10, IL-21, tumor necrosis factor (TNF)-α, lysozyme, Toll-like receptors (TLR)-1, TLR-3, and TLR-4 were significantly upregulated in zebrafish body; apoptosis-related genes such as superoxide dismutase (SOD)1, glutathione peroxidase (gpx)1α, and B-cell leukemia (bcl)2 were upregulated, downregulated expression level of nitric oxide synthase (nos)2α and tumor protein (tp)53 in zebrafish liver after feeding with amy wild type and amy R8. phagocytic activity, respiratory burst, and lysozyme activity were enhanced in tilapia after feeding with amy wild type and amy R8 for 2 months. Increasing of hepatic metabolism genes such as hepatic glucokinase (GK), glucose-6-phosphate dehydrogenase (G6PD), glucose-6-phospate (G6Pase), and insulin growth factor-1 (IGF-1) and decreasing of antioxidant related genes such SOD and catalase (CAT) were found in tilapia after feeding with amy wild type and amy R8. In conclusion, our results suggested that dietary supplementation of B. amyloliquefaciens (amy wild type) and xylanaseexpressed B. amyloliquefaciens (amy R8) promoted growth performance in tilapia, enhanced innate immune and hepatic stress ability in increase of disease resistance against pathogens in zebrafish and tilapia.
