Abstract
Penurunan apoptosis limfosit merupakan salah satu faktor penyebab keradangan kronis saluran nafas pada asma alergik. Beberapa penelitian menunjukkan peran interleukin (IL)-4 dalam penghambatan apoptosis limfosit, namun masih sedikit penelitian tentang peran Bax-limfosit dan hubungannya dengan IL-4 pada asma. Tujuan penelitian ini adalah membuktikan peran IL-4 dalam menghambat apoptosis limfosit melalui penurunan Bax-limfosit pada bronkiolus dan paru mencit asma. Penelitian ini merupakan penelitian eksperimental dengan metode randomized control group . Subyek penelitian adalah mencit Balb/c yang dibagi dalam 2 kelompok: asma dan non-asma. Kelompok asma disensitisasi dengan ovalbumin secara intraperitoneal pada hari ke-0 dan 14, diikuti dengan cara inhalasi setiap 2-3 hari selama 6 minggu. Pada minggu ke-8, seluruh subyek diterminasi. Ekspresi IL-4 dan Bax-limfosit diperiksa dengan metode imunohistokimia, sedangkan apoptosis limfosit dengan metode TUNEL. Analisis statistik menggunakan independent sample t-test , Mann-Whitney U test, analisis regresi, dan analisis jalur dengan interval kepercayaan 95%. Spesimen bronkiolus dan paru didapatkan dari 18 subyek (9 dari masing-masing kelompok). Jumlah apoptosis limfosit sama antara kedua kelompok ( p =0,116), sedangkan Bax-limfosit menurun pada kelompok asma ( p =0,003). Hal ini menunjukkan penurunan apoptosis limfosit pada kelompok asma. Sebaliknya, ekspresi IL-4 meningkat pada kelompok asma ( p =0,00). Terdapat hubungan negatif sedang ( r 2 =0,29, p =0,011) antara IL-4 dan apoptosis limfosit. Terdapat pula hubungan negatif kuat ( r 2 =0,39, p =0,002) antara IL-4 dan ekspresi Baxlimfosit. Namun, tidak ditemukan hubungan antara Bax-limfosit dan apoptosis limfosit ( r =0,37, p =0,065). Analisis jalur menunjukkan pengaruh IL-4 terhadap apoptosis limfosit secara langsung lebih besar dibandingkan pengaruh tidak langsung melalui penurunan Bax limfosit. Sebagai kesimpulan, peningkatan ekspresi IL-4 menghambat apoptosis limfosit, namun bukan melalui penurunan Bax-limfosit.
English Abstract
Decrease of lymphocyte apoptosis is one factor that leads to chronic airway inflammation in allergic asthma. Previous studies revealed role of interleukin (IL)-4 in preventing lymphocyte apoptosis; however re are few studies about role of lymphocyte-Bax and its relationship with IL-4 in asthma. aim of study is to demonstrate IL-4 role in preventing lymphocyte apoptosis via lymphocyte-Bax inactivation in bronchioli and lungs of asthmatic mice. It is a randomized control group experimental study. subject was Balb/c mice that categorized into 2 groups: asthma and non-asthma. Asthma group was sensitized by ovalbumin intraperitoneally in day 0 and 14, followed by inhalation every 2-3 days for 6 weeks. In week 8, all of mice were terminated. IL-4 and lymphocyte-Bax expression was examined through immunohistochemistry method, whereas lymphocyte apoptosis by TUNEL method. Independent sample t-test, Mann Whitney U test, regression analysis, and path analysis was used in statistical analysis with confident interval 95%. bronchioli and lungs specimens were obtained from 18 mice (9 in each group). Lymphocyte apoptosis was similar between 2 groups ( p =0.116), additionally lymphocyte-Bax decreased in asthmatic group ( p =0.003). This indicated low activity of lymphocyte apoptosis in asthmatic group. Conversely, IL-4 expression increased in asthmatic group ( p =0.00). re was moderate negative correlation ( r 2 =0.29, p =0.011) between IL-4 and lymphocyte apoptosis. re was also strong negative correlation ( r 2 =0.39, p =0.002) between IL-4 and lymphocyte-Bax expression. However, we did not find any correlation between lymphocyte-Bax and lymphocyte apoptosis ( r =0.37, p =0.065). Path analysis revealed stronger direct relationship between IL-4 and lymphocyte apoptosis ra r than indirect relationship via lymphocyte-Bax inactivation. We conclude that increases of IL-4 expressions inhibit lymphocyte apoptosis; however, it is not due to Bax-lymphocyte inactivation.
