Abstract

Tujuan: Penelitian ini bertujuan untuk membuktikan pengaruh pemberian genistein dosis tinggi terhadap ekspresi protein Ki-67 pada kultur sel endometriosis. Metode: Suatu studi eksperimental yang dilakukan di Laboratorium Fisiologi Fakultas Kedokteran Universitas Brawijaya Malang dengan menggunakan kultur sel endometriosis. Jaringan endometriosis diambil dari pasien penderita endometriosis melalui tindakan laparoskopi, dilakukan kultur hingga konfluent. Kultur dibagi menjadi 7 kelompok perlakuan yaitu kultur tanpa perlakuan, kultur + genistein 5μM/L, kultur + genistein 10 μM/L, kultur + genistein 20 μM/L, kultur + genistein 30 μM/L, kultur + genistein 40 μM/L, kultur + genistein 50 μM/L, kemudian di inkubasi pada suhu 37˚C selama 6 jam, 24 jam dan 48 jam. Ekspresi protein Ki-67 diketahui dengan pemeriksaan flowcytometri. Data hasil pengamatan dianalisis dengan uji ANOVA dan uji regresi. Hasil: Genistein mampu menurunkan ekspresi protein Ki-67 terendah pada pemberian dosis 40 μM/L (30.23+11.82 %) dan 50 μM/L (30.49+16.73 %) bila dibandingkan dengan perlakuan dosis yang lain. Terdapat perbedan bermakna rerata ekspresi Ki-67 antara kelompok waktu inkubasi 6 jam (12.08+2.74 %), 24 jam (45.03+4.08 %), dan 48 jam (38.88 + 2.87 %). Kesimpulan: Adapengaruh yang bermakna pemberian genistein dosis tinggi terhadap penurunan ekspresi protein Ki-67 pada kultur sel endometriosis, terutama pada dosis 50 μM/L pada waktu inkubasi 6 jam.

English Abstract

Objective:To prove effect of high doses of genistein on expression Ki-67 protein in cell culture endometriosis. Methode: This study had been performed in Physiology Laboratorium, Medical Faculty of Brawijaya University Malang, using cell culture endometriosis.Endometriosis tissue collected from patients with endometriosis via laparoscopy, was cultured to make a confluent culture. culture was divided into 7 treatment groups, culture without treatment, culture + genistein 5μM / L, culture + genistein 10μM / L, culture + genistein 20μM / L, culture + genistein 30μM / L, culture + genistein 40μM / L, culture + genistein 50μM / L, and n incubated at 37 ˚ C for 6 hours, 24 hours and 48 hours. Ki-67 protein expression was determined by examination flowcytometri. data were analyzed with ANOVA and regression test. Result: Genistein is able to decrease expression of Ki-67 protein in lowest dose 40 μM / L (30.23 +11.82%) and 50 μM / L (30.49 +16.73%) when compared with o r treatment doses. re was significant expression of Ki-67 between incubation time of 6 hours (12.08 +2.74%), 24 hours (45.03 + 4.08%), and 48 hours (38.88 + 2.87%). Conclution: re was significant effect on administration of high doses of genistein to decrease expression of Ki-67 protein in cell culture endometriosis, especially at doses of 50 μM/L at 6 hours incubation time.
