Abstract

Tujuan : Penelitian ini bertujuan untuk mengidentifikasi pengaruh genistein pada kultur sel endometriosis dalam menurunkan kadar IL-1β. Metode : Penelitian dilakukan secara eksperimental laboratorik di laboratorium Fisiologi Fakultas Kedokteran Universitas Brawijaya Malang menggunakan posttest only control group design. Sampel pada penelitian ini adalah jaringan endometriosis hasil dari prosedur laparoskopi. Kultur sel endometriosis dibagi menjadi 7 kelompok perlakuan (kelompok kontrol, kelompok dosis 5 μmol/L, dosis 10 μmol/L, dosis 20 μmol/L, dosis 30 μmol/L, dosis 40 μmol/L dan dosis 50 μmol/L) dan 3 waktu inkubasi ( 6 jam, 24 jam, 48 jam). Kadar IL-1β diukur dengan menggunakan enzyme-linked immunosorbent assay (ELISA). Data dianalisis secara statistik dengan uji ANOVA dan uji regresi. Hasil : Pemberian genestein berbagai dosis dan waktu inkubasi berpengaruh terhadap penurunan kadar IL-1β (p = 0.000). Kadar IL-1β terendah didapatkan pada kelompok dosis 50 μmol/L inkubasi 48 jam (112.98±6.42 pg/ml), namun secara statistik tidak berbeda signifikan dengan kelompok dosis 40 μmol/L. Kesimpulan : Pemberian genistein berbagai dosis dan waktu inkubasi berpengaruh terhadap penurunan kadar IL-1β pada kultur sel endometriosis. Semakin tinggi dosis genistein dan semakin lama waktu inkubasi, kadar IL-1β pada kultur sel endometriosis cenderung menurun Kata kunci: Endometriosis, Genistein, IL-1β

English Abstract

Objective : Identification effect of genistein on endometriosis cell culture in lowering level of interleukin-1β (IL-1β). Method : study was conducted experimental laboratory with posttest only control group design in Physiology Laboratory Faculty of Medicine, Brawijaya University Malang. Isolated tissue sample obtained from endometriosis patient through laparoscopy procedure. Endometriosis cell culture divided into 7 treatment groups (control group, genestein dose 5 μmol/L, 10 μmol/L, 20 μmol/L, 30 μmol/L, 40 μmol/L and 50 μmol/L) with an incubation period of each dose (6 hours , 24 hours and 48 hours). IL-1β concentrations were assessed by using commercial enzyme-linked immunosorbent assay (ELISA). Data were analyzed with ANOVA and regression test. Result : Administration of various doses of genistein and incubation time had effect on decreased level of IL-1β (p = 0.000). lowest levels of IL-1β on dose 50 μmol/L with 48 hours incubation (112.98±6.42 pg/ml), but did not statistically significant different compare with dose 40 μmol/L. Conclusion : Administration of various dose of genistein and incubation time had effect on decreased level of IL-1β in endometriosis cell culture. higher dose of genistein and longer incubation time, levels of IL-1β is increasingly affecting tend to decreased.
