Abstract

Tujuan: Penelitian ini bertujuan untuk membuktikan pemberian genistein dapat menurunkan kadar MMP-2 pada kultur sel endometriosis Metode: Penelitian ini merupakan penelitian eksperimental yang dilakukan di Laboratorium Fisiologi Fakultas Kedokteran Universitas Brawijaya. Menggunakan jaringan endometriosis yang didapatkan dari pasien penderita endometriosis melalui tindakan laparoskopi, kemudian dilakukan proses kultur hingga konfluent. Kultur dibagi menjadi 7 kelompok, yaitu : kelompok tanpa perlakuan, pemberian genistein dosis 5μM/L, 10μM/L, 20μM/L, 30μM/L, 40μM/L, 50μM/L dengan waktu inkubasi 6 jam, 24 jam, 48 jam. Pengukuran kadar MMP-2 menggunakan ELISA kit. Data hasil pengamatan dianalisis dengan uji ANOVA dan uji regresi. Hasil: Ada pengaruh pemberian genistein terhadap kadar MMP-2 pada kultur sel endometriosis (p 0,05).Genistein mampu menurunkan kadar MMP-2 terendah pada dosis 30μM/L (3.57±1.94ng/ml), 40μM/L (3.44±1.47ng/ml) dan 50μM/L (3.54±1.3d ng/ml) bila dibandingkan dengan perlakuan dosis yang lain. Rata-rata kadar MMP-2 pada waktu inkubasi 6 jam (2.21±0.93a ng/ml)lebih kecil daripada waktu inkubasi 24 jam (3.81±0.91 ng/ml) dan 48 jam (7.94±3.08 ng/ml). Kesimpulan: Genistein dapat menurunkan kadar MMP-2 pada kultur sel endometriosis, terutama pada dosis 30 μM/L dan waktu inkubasi 48 jam.

English Abstract

Objective: To determine effect of genistein to decrease MMP-2 levels in endometriosis cell culture. Methode: This design of this study was experimental, had been performed in Physiology Laboratory Medical Faculty of Brawijaya university. This study used endometriosis tissue from patients with endometriosis via laparoscopy, cultured process to make aconfluent. culture was divided into 7 groups: untreated group, administration genistein dose of 5μM / L, 10μM / L, 20μM / L, 30μM / L, 40μM / L, 50μM / L with incubation time 6 hours, 24 hours, 48 hours. measurement of MMP-2 using ELISA kits. data were analyzed with ANOVA and regression test. Result: re was effect on administration genistein to MMP-2 levels in endometriosis cell culture (p 0.05).Genistein is able to decrease MMP-2 level in lowest dose 30μM/L (3.57 ± 1.94 ng / ml), 40μM/L (3.44 ± 1.47 ng / ml) and 50μM / L (3.54 ± 1.3d ng / ml) when compared with o r treatment dose. average of MMP-2 levels at 6 hours incubation time (0.93 ± 2.21 ng/ml) is smaller than 24-hour incubation time (3.81 ± 0.91 ng/ml) and 48 hours (7.94 ± 3.08 ng/ml). Conclusion: Genistein was decrease MMP-2 levels in endometriosis cell culture, especially at dose 30 μM/L and an incubation time of 48 hours.
